Objective: Traumatic osteomyelitis bone disease treatment is hot and difficult, with a costly, difficult to treat, duration of delayed healing, and high morbidity. More common in high-energy injury caused open fractures, lesions often accompanied by severe soft tissue damage. Currently in the treatment of a variety of methods to remove infected bone though, it was still difficult to solve fibrosis patients infected with the surrounding muscle tissue. Surrounding soft tissue fibrosis caused by stiff joints often leads to loss of limb function. How can the soft tissue left over after treatment of osteomyelitis caused by excessive fibrosis gradually clinicians importance. By pathological examination, Masson staining, immunohistochemical analysis, Western Blot and experimental methods to observe Decorin in rat skeletal muscle hypertrophic scar tissue TGF-?1, phosphp-Smad3 and collagen type I cytokine expression, and to explore the mechanism and experimental study of Decorin suppressing scar osteomyelitis model in rats.Method: The modified design of the establishment Contusion modeling method in the rat model of chronic osteomyelitis, the 90 healthy and clean male(Wistar) rats were divided into three groups(n = 30): group A(bacterial concentration 106 CFU / ml), group B( PBS intervention)and group C. Decorin intervention group was given the day after 10 ug / ml Decorin dilution local intramuscular injection once every five days later, PBS intervention group were given injections of local PBS measured, osteomyelitis, non-intervention group. Taking tissue using pathological examination, Masson staining, immunohistochemistry, Western blotting and immunohistochemistry(Western blot) Evaluation of chronic osteomyelitis in skeletal muscle scarring degree after 1th, 2th and 4th weeks.Result: 1.(1) After 4 weeks, group C was found by Masson staining of collagen fibers arranged in neat, and significantly less than A, B groups; no significant difference between group A and group B.(2) Immunohistochemical staining, TGF-?1 expression of collagen type I and group C was significantly lower than group A, B; A, B two groups did not differ significantly.(3) Western blot show, TGF-?1 expression of collagen type I and group C was significantly lower than A, B groups, with significant difference(p <0.01); A group of TGF-?1 and collagen type I expression was significantly lower than B group, no significant difference(p> 0.01).2. Group C compared with the group A by HE staining observed after 2 weeks and 4 weeks significantly reduced the degree of fibrosis, Western blot showed that TGF-?1, phosphp-Smad3 and type I collagen levels were significantly reduced, no significant difference after 1 weeks.Conclusion: 1. Decorin can effectively suppress traumatic osteomyelitis caused by skeletal muscle fibrosis.2. PBS had no effect on the inhibition of skeletal muscle fibrosis, osteomyelitis.3. Decorin inhibit scarring in rat skeletal muscle osteomyelitis extent, this process occurs with the TGF-?- Smad signal pathway.
