To Investigate The Effects Of Erzhi Tiangui Granules On Ovarian Granulosa Apoptosis By C-FLIP Factor

Objective: Based on c-FLIP factor taking the effect of Erzhi Tiangui Granules on ovarian granulosa apoptosis and egg cell quality of QI-deficiency type of the kidney patient, who is carried out in vitro fertilization and embryo transplantation(IVF-ET).Methods: 70 senile(35~42) infertile patients with tubal factors undergoing IVF-ET were divided into two groups at random, 35 cases in the observation group treated with Erzhi Tiangui Granules for three menstrual cycles, and 35 cases in the placebo group were treated with placebo for three menstrual cycles.Then two groups were carried out controlled ovarian hyperstimulation short program treatment after above treatment. To observe the clinical basic situation and ovarian reserve of two groups of patients before treatment, kidney symptom scores were compared between before and after treatment, the day of HCG serum E2 ?P?LH level, the dose of gonadotropin(Gn), Treatment time,the number of oocytes retrieved, the rates of fertilization, high quality oocytes rate, high quality embryo rate, clinical pregnancy rate and c-FLIP, Caspase 3 protein expression levels in ovarian granulose cells.Results:(1)After treatment, the observation group patients in the clinical symptoms(integration) improvement was superior to the placebo group(p <0.01), the difference was statistically significant.(2) Observation group patients were superior to placebo group(p <0.05) in the dose of Gn, treatment time and the day of hCG serum E2 / P levels, the difference was statistically significant.(3) Observation group patients were superior to placebo group(p <0.05)in the high quality oocytes rate, high quality embryo rate,the difference was statistically significant.Number of oocytes retrieved, fertilization rate was no significant difference(P <0.05).(4) Observation group patients were superior to placebo group(p <0.05) in the clinical pregnancy rate, the difference was statistically significant. Biochemical pregnancy rate was no significant difference(P <0.05).(5) Observation group patients were superior to placebo group(p <0.01)in c-FLIP, Caspase 3 protein expression levels in their ovarian granulose cells.Conclusions:(1)Application of IVF-ET treatment with Erzhi Tiangui Granules can significantly improve senile infertile patients with Qi-deficiency type of the kidney patient,increasing the sensitivity of oocytes of exogenous Gn, the dose of gonadotropin(Gn), Treatment time,the number of oocytes retrieved, the rates of fertilization, high quality oocytes rate, high quality embryo rate, clinical pregnancy rate.(2)Erzhi Tiangui Granules can suppress Fas-mediated apoptosis pathway of granulosa cells by c-FLIP factor.It can inhibit the apoptosis of granulosa cells and promote the normal growth of the oocyte follicle development, maturation and ovulation.Improving oocyte quality and improving sealing possession of ovarian function to avoid non-ovarian sometimes vent.This may be one of the mechanisms to improve IVF pregnancy outcome of kidney traditional Chinese medicine.This research provide a more objective basis for the "main reproductive kidney" theory from the protein levels of ovarian granulosa cell apoptosis.
