Study On Clinical Intervention Of Tangyisheng-1 Decoction On T2DM And Experimental Study On GLP-1 Of Rat

Objective:To investigate the clinical effects and the mechanism of Tangyisheng-1 decoction which could remove heat and toxicant, invigorate the spleen and kidney, disperse lung and bowel, unobstruct triple energize on moderate and severe T2 DM.Methods:1.Clinical Research: The selected 70 cases patients with severe hyperglycemia in Type 2 diabetes mellitus were randomly divided into treatment group(35 cases)and control group(35 cases).The two groups were based on the foundation treatment, and then were given Tangyisheng-1 decoction combined with metformin(Glucophage)and metformin(Glucophage) in 12 weeks for a course of treatment. Observe the main symptoms and physical signs and abnormal physical changes in the index of the two groups before and after treatment, and then compare.2.Experimental Research: The diabetic model rats were made with STZ.40 male Wistar rats in successful model were randomly divided into 4 groups as model control group, metformin group, the low-dose Tangyisheng-1 decoction group, the high-dose Tangyisheng-1 decoction group, and 10 rats in each group. Another 10 normal rats were taken as blank control group. After 6 weeks of drug administration, the portal GLP-1 was detected by enzyme linked immune method and the expression of GLP-1 in the small intestine and colon was detected by using immunohist ochemistry method.Results:Clinical Research:Tangyisheng-1 decoction combined with metformin have obviously improved in clinical symptoms and signs, as well as significant effect in lowering blood glucose,HbA1 c,increasing C peptide and insulin release and regulating blood lipid better than the control group.2. Experimental Study:Tangyisheng-1 decoction not only can obviously improve the fasting blood glucose in diabetic rats, weight loss, and promote the secretion of insulin but also can significantly enhance the secretion of GLP-1.The effect of the high-dose Tangyisheng-1 decoction group is better than that of the control groups which suggests that Tangyisheng-1 decoction has a good effect on regulating the abnormal glucose metabolism.Conclusion: The effect of Tangyisheng-1 decoction which could remove heat and toxicant,invigorate the spleen and kidney,disperse lung and bowel,unobstruct triple energize on T2 DM is exact.The mechanism may be related to improve glucose and lipid metabolism,apoptosis factors and it is worth further research.
