| Aims:Establishment of rat carotid artery after balloon injury model, to investigate the early preventive effects of Onychin and its mechanism on vascular intimal hyperplasia by observing the effect of Onychin on the common carotid artery intimal hyperplasia after balloon injure in aortic arteries of rats.Methods:Sixty-four male SD rats(weight of about 300-400g) were randomly divided intosham-operated group,model group,HDG(Onychin high dose group) and LDG(Onychin low dose group).The rats in the two Onychin groups were given Onychin 20 mg/kg or 80 mg/kg daily from the week before surgery,respectively.The rats in the model group and the two Onychin groups were induced neointimal hyperplasia by carotid artery balloon injury.After the rats were sacrificed at day 7 and 14,HE staining was used to observe the change of vascular morphology and the thickness of the arterial wall. Immunohistochemistry staining and Western Blot were performed to measure the expression of p ERP1/2.Results :1. The morphological changes of endangium were studies with HE staining:On the 7th and 14 th day after operation,compared with the sham-operated group, the thickness of vessel wall increased significantly in the other three groups;compared with the model group,the vessel wall thickness were significantly decreased in the two Onychin groups. 2.The expression of p ERK1/2 detected by the method of immunohistochemical staining and Western Blot: compared with the sham-operated group, the expression of p ERK1/2 increased obviously in the other three groups in postoperative 14 days,explain that the balloon injury could increase the expression of p ERK1/2( p <0.05); the expression of p ERK1/2 reduced significant in the Onychin group(p <0.05).Conclusion:1.Onychin can inhibit the proliferation of vascular intima after carotid balloon injury in rats as well as dose-dependent. 2.Onychin attenuates neointimal hyperplasia and angiostenosis in the rat carotid artery balloon injury model, which may be related to down-regulated expression of p ERK1/2. |
