Objective:The study was aimed to investigate the growth and apoptosis in human kidney cancer ACHN cells induced by DADS, and to observe the protein of Akt and Livin expression in the cells after DADS treatment to human kidney cancer ACHN cells. To find out the mechanism, and provide the theoretical basis for treatment human kidney cancer.Methods:Kidney cancer ACHN cells were treated at different concentrations in vitro. The effect of DADS on the proliferation ability of the cell was measured by MTT assay. Apoptosis rate and cell cycle distribution were analyzed by flow cytometry(FCM). The expression of Akt and Livin were detected by Western blotting analysis.Results:1. DADS inhibition of kidney cancer ACHN cellsMTT results showed that human kidney cancer ACHN cells were inhibited by DADS.Different concentrations of DADS intervention after kidney cancer ACHN cells at different times,the value of OD570 was lower than the control group.With the increase of concentration and time,the OD570 value was significantly lower, and its inhibitory effect on the kidney cancer ACHN cells increase concentration and time dependence(P<0.05).2. DADS can induce kidney ACHN cells apoptosisFlow cytometry analysis showed that concentration of 10ug/ml, 20ug/ml and 40ug/ml DADS 48 h after intervention in kidney cancer ACHN cells respectively, apoptosis rate was lower than the control group,and its enhanced effect on the kidney cancer ACHN cells increase concentration dependence(P<0.05).3.The influence of DADS on expression of AKT and Livin protein in kidney cancer ACHN cells.Western Blotting demonstrated that after oncentration of 20ug/ml DADS after intervention in kidney cancer ACHN cells,the expression levels of AKT and Livin were significantly lower in the DADS infected cells than the control group(P<0.05).Conclusion:1.DADS can inhibit the growth of human kidney cancer ACHN cells,and its inhibtion rate was time,concentration-dependence increase.2.DADS can induce kidney ACHN cells apoptosis,may be associated with deccreased expression of AKT and Livin.
