Objectives The aim of this study was based on the model of surgical stress to confirm that sevoflurane has effect on stress reaction in the process of operation to restrain the stress and inhibit the function of TASK1/3 potassium channels.Methods There are 32 C57BL/6 mice selected respectively of wild type TASK1+/+?TASK3+/+ and knockout type TASK1-/-?TASK3-/-. A total of 64 adult C57BL/6 mice(male and female),age 6 to 8 weeks,weight 18-25 g,were randomly divided into 8 groups,each group of eight(n=8).Wild type TASK1+/+?TASK3+/+ can be divided into 4 groups:blank group(C1), sevoflurane group(Q1), surgical group(S1),surgery+sevoflurane group(H1).Knockout type TASK1-/-?TASK3-/- also can be divided into 4 groups:blank group(C2), sevoflurane group(Q2),surgical group(S2),surgery+sevoflurane group(H2).All mice were intraperitoneal injected preoperatively of 1% pentobarbital 100mg/kg.After being anesthesia,all mice have be accomplished with preoperative based life monitoring:including blood glucose,heart rate,blood pressure.The task of group Q1 and Q2 were accepted pure sevoflurane inhalation(2.5%,Oxygen Flow 2L/min) after basal anesthesia,group S1 and S2 were used the animal model of abdominal surgery after basal anesthesia,the group H1 and H2 were accepted sevoflurane inhalation(2.5%,Oxygen Flow 2L/min) and abdominal surgery at the protein and DNA expression of TASK1/3 potassium channels in adrenal of mice with type of TASK1+/+,TASK3+/+and TASK1-/-,TASK3-/-. the same time.Then,experimental record includes blood glucose,heart rate and blood pressure.Blood was sampled from mice eyes and using high-performance liquid chromatography(HPLC) method for the determination of plasma norepinephrine(NE) and epinephrine(E) concentration.Using enzyme-linked immunosorbent assay(ELISA) method for determination of plasma corticosterone(COR) concentration.Using Western Blot and PCR method to testResults According the Western Blot and PCR results show that TASK1/3 potassium channels have been expressed in adrenal tissue of TASK1+/+?TASK3+/+ but not in type of TASK1-/-?TASK3-/-.The surgical stress response in mice blood pressure,heart rate,content of plasma catecholamine and corticosterone show that the type of TASK1-/-,TASK3-/- are higher than the type of TASK1+/+,TASK3+/+(P<0.05).After sevoflurane inhalation anesthesia,potency of sevoflurane was lower in the type of TASK1-/-,TASK3-/-. TASK1-/-,TASK3-/- type and TASK1+/+,TASK3+/+ type in blood glucose have no significant difference(P>0.05).Conclusions The mechanism of Sevoflurane restrain surgical stress response could be by activating the adrenal TASK1/3 potassium channels,and the TASK1/3 potassium channels could also be action targets of Sevoflurane.
