Objective:Based on the previous study of differentiation from rat Bone marrow mesenchymal stem cells(BMSCs) to neuron-like cells with the induction of Mecobalamin in vitro, a further study was made on whether the BMSCs of the rats with local injection and induction of Mecobalamin can achieve the differentiation into neuron-like cells, as well as on the recovery effect of spine cord injuries that the differentiated neuron-like cells contribute to. In the present study, experiments of animal models were conducted to study the treatment for Spine cord injuries on Mecobalamin combined with BMSCs transplantation, in order to discuss the feasibility of the differentiation of BMSCs into neuron-like cells, and to provide theoretical and experimental basis for the new treatment of Spine cord injuries.Method:Divide 40 SD female rats which were taken as T10 spinal cord transection models into four groups at random, into four groups at random. Each group includes 10 rats. So there are group A(the model control group), group B(Mecobalamin treatment group), group C(BMSCs transplantation group) and group D(Mecobalamin and BMSCs combined transplantation group),. On the 1st, 7th, 14 th and 21 st days after surgery,make BBB scores on the lower limbs, have HE dyeing observation of white and grey matter of spinal cord and the damage condition of the intercellular substance, and conduct a Nissl staining to observe the state and distribution of the survival neurons as well as the condition of their regeneration.Results :1. After molding, all the four groups of rats had serious dysfunction of lower limbs. And comparing the BBB score of the four groups, we found that the differences make no statistical sense(p<0.05). During the7 th to 21 st days after surgery, the BBB score of each group was getting higher as time went by, which means after spinal cord injury rats got recovery in some degree, with group C and D better than C, group B better than A.2.Spinal cord tissue pathological section showed that group A got the most serious spinal cord injury, with neuron structure in disorder, a large amount of inflammatory cell infiltration, and slow reparation process with a low degree. Group B and C were better than group A. It also showed that group D got slightly tissues injured and faster reparation process. Under the immunofluorescent microscope, it can be observed that there were some neuron-like cells with fluorescence in both group C and group D, with the fluorescence intensity and density of group C less than group D. Among all the Nissl stained sections, the amount of remained Nissl body as well as that of the regenerating Nissl body in group D is the biggest.conclusions:1. Bone marrow stromal cells transplant and Mecobalamin treatment can improve the rat with embryonic stem cells motor function of lower limbs, and combining the two methods of treatment can gain better result.2. Bone marrow stromal cells can be divided into neuroepithelial stem cell in the environment of myoloid tissue, and Mecobalamin can help promote the division of BMSCs to neuroepithelial.3.Combining the Mecobalamin and BMSCs transplant can relive the pathological change of embryonic stem cells.
