Research Background:The central nerve regeneration has been a confused problem in the theory research and clinal practice of neurosciences area, however, there is no effective method to solve the problem.In developed country, the incidence of spinal cord injury is about13.3to45.9persons per million one year.There is no statistical material for this in our country, and according to rough estimate, about10thousand persons will get the spinal cord injury one year.The spinal cord injury will cause the waste of labor force,and as the same time it will bring hard burden for the family and society.Recently, with the development of cell engineering, cell transplantion therapy has been a hot point to investigate the spinal cord injury.Neural stem cell, fibroblast, olfactory ensheathing cells, macrophage has been report to be efficenicy to repair the spinal cord injury. However, it can not meet our demond. So finding the new and efficiency cell to treat spinal cord injury has been the import way to get the breakthrough in this aera. Since the bone marrow mesenchymal stem cells (BMSCs) were recowered, lots of datas have been demonstrate that the BMSCs has the potential to differentiate to bone, cartilage, skeletal muscle,fat and ligament.Kopen reports that BMSCs transplanted to tricorn of neonatal rat can differentiate to neuroglia,and BMSCs begin to be used to treat the spinal cord injury. However, curative effect of pure BMSCs transplantation to treat spinal cord injury is not good enough. Abouelfetouh use neural cells differentiated from bone marrow mesenchymal stem cells (BMSCs) by the co-culture with hippocampus cells to treat the spinal cord injury.In our research, we use biology and chemisty method to induce the BMSCs to differentiate to neural cells, and transplant the neurocells to the prepared SCI model. By the comparison of the behavior and detection of the biochemical criterion of the normal rats and spinal cord injuried rats, we study the curative effect of neural cells differentiated from bone marrow mesenchymal stem cells (BMSCs) transplantation on the recovery of rats with spinal cord injury. The experiment provide the theory evidency to spport the therapy of BMSCs transplantion.Objective:The curative effect of neuron-likel cells differentiated from bone marrow mesenchymal stem cells (BMSCs) transplantation on the recovery of rats with spinal cord injury (SCI) was investigated by the comparison of the behavior and detection of the biochemical criterion of the normal rats and spinal cord injuried ratsMethods:A4-week old mouse was utilized to separate BMSCs which were cultured into third generation and then chemically-inducement and molecule inducement was used to induce the differentiation of BMSCs.488-weeks old male wistar rats were used to establish the SCI model with Allen’s method and divided into group A, group B, group C and group D randomly. Rats in group A were injected the third generational purified BMSCs, rats in group B were injected with the chemically-induced BMSCs, rats in group C were injected the molecules-induced BMSCs, rats in group d were injected with the cell culture medium. Basso-Beattle-Bresnahan(BBB) were observed at the first, second, third, fourth, sixth, eighth, tent week and twelfth week after the establishment of the SCI model. At the weekend of the twelfth week, we use the injured part of spiral cord to make the tissue slice and then observe the repairmen of the injured spinal cord.Results: Afer the twelve week since we establish the SCI model, the recovery of posterior limb’s function of rats in group A, group B and group C is significantly better than that in group D (P<0.0001). There was no significant difference of the BBB between the group A and group B(P=0.4363), and group C is better than that in group A(P<0.05) and group B(P<0.05). The HE staining of spinal cord tissue slice showed that astrocytes-spinal cord from rats of group A,group B and group C is more proliferative than that from group D, And the disintegration of nerve cell and spinal cord stat were less than that in group D. The recovery effect of rat from group C is the best of the Four groups.Conclusion:1. Both the biology and chemisty inducing method can induce the BMSCs to differentiate to neuron-like cells.2. The transplantion of BMSCs and neuron-like cells differentiated from BMSCs with two inducing method can ameliorate the spinal cord injury.3. The transplantion of neuron-like cells differentiated from BMSCs with the biology inducing method has better curative effect than transplantion of BMSCs and neurocells induced by chemisty method.
