The Influence Of Jianpi Qinghua Decoction For The Patients With Acute-on-chronic Liver Failure In Vitro T Lymphocytes And Cytokines

Objective: The purpose of this paper is to observe The influence of Jianpi Qinghua Decoction for the patients with damp heat type of HBV related acute-on-chronic liver failure in vitro T lymphocyte cells differentiation and the changes of cell supernatant TNF-?,IFN-?,IL-18,IL-10 levels,in order to investigate the immune mechanism of Jianpi Qinghua Decoction on the patients with damp heat type of HBV related acute-on-chronic liver failure,thus providing theoretical basis for traditional Chinese medicine treatment of liver failure.Methods: We extracted the peripheral blood mononuclear cells of 20 cases with damp heat type of HBV related acute-on-chronic liver failure,extracted the lymphocyte cells after stick wall cell culture.Culture the T cells with Jianpi Qinghua Decoction in rats serum and blank control group in vitro respectively,and collected the lymphocyte cells and cell supernatant.Using flow cytometry instrument to detect the changes of T cell subgroup,ELISA quantitative detect the cells supernatant TNF-?,IFN-?,IL-18,IL-10 level changes.Compare the differences of the two grops in immune cells and cytokines.Results:1.After medicated serum stimulates the cells,the percentage of CD3+?CD4+ were higher than blank control group,percentage of CD8+ were lower than blank control group,and CD4+/CD8+ rate were higher than blank control group,the difference has statistically significant(P = 0.000<0.05).2.After medicated serum stimulates the cells,the levels of TNF-??IFN-??IL-18 in cells supernatant showed a downward trend,the levels of IL-10 in cells supernatant were on the rise.Comparison of cells supernatant TNF-??IFN-??IL-18 ?IL-10 levels between two groups significantly different(P = 0.000<0.05).Conclusion: 1.Jianpi Qinghua Decoction could increase the rate of CD3+,CD4+ and CD4 +/CD8+ rate and decrease the percentage of CD8+,and improve T cell subsets disorder status.This prompted that Modified Jianpi Qinghua Decoction could regulate the overall cellular immune imbalance of patients.2.Jianpi Qinghua Decoction could reduce the levels of TNF-?,IFN-?and IL-18 in acute-on-chronic liver failure patients' lymphocyte cells supernatant and increase levels of IL-10.This prompted that Jianpi Qinghua Decoction might by improving the Th1/Th2 imbalance,and reduce the degree of liver damage,delay the progression of the disease.
