Objective:Build two types of animal models of allergic rhinitis and asthenia cold sysdrome on guinea pigs to study the effects on guinea pigs with allergic rhinitis disease and syndrome chang by separate components for Mahuangfuzixixin decoction.Form the point of view that the transcript-ion factor GATA-3 controls Th1 and Th2 generation to explore the compatibility of prescription rationality and mechanism.Evaluation the clinical efficacy of the jiaweimahuangfuzixixin decoction in treating asthenia cold sysdrome of allergic rhinitis.Methods:1. Animal experiment:Make 200%decoction to gavage by the proportion of Shigao:Zhimu:Huangbai:Longdancao=1.5?2.0?1.0?1.2,and then use fumigation to build the model of asthenia cold sysdrome. When completed,use intranasal method to build the model of AR. 3-month-old cleaning stage guinea pigs were randomly divided into six groups, they are the group of mahuangfuzixixin,mahuangfuzi,mahuangxixin,mahuang,Dexamethasone and model group. Record the change of symptoms and signs, detecting GATA-3 expression of nasal mucosa of guinea pigs by immunohistochemistry method after sacrificed.2. Clinical Observation:Select 30 cases of allergic rhinitis patients who meet the inclusion criteria, giving them the treatment of taking jiaweimahuangfuzixixin decoction. 4 weeks for a course of treatment, continu-ous observation of a course. Record the change of symptoms and signs before and after treatment.Results:1. Animal experiment:(1) The measurement results of guinea pig OHCS-17 and F: After the experiment, all the experimental groups are high expression compared to after modeling, and there existing significant difference(p<0.05) with the model group. Among them,the group of mahuangfuzixixin and mahuangfuzi, dexamethasone are highly expressed, comparison with the rest of the group the difference are significant(p <0.05). After the experiment,except for the group of mahuang,rest of the experimental group are high expression compared to after modeling, and existing a significant difference(p<0.05) with the model group. Among them,the group of mahuangfuzixixin,mahuangfuzi,mahuangxixin and dexamethasone are highly expressed, comparison with the rest of the group the difference are significant(p<0.05).(2) Guinea pig nasal GATA-3 immunohistochemical results in each group after the experiment : semiquantitative statistics GATA-3 expression(staining index + percentage of positive cells), GATA-3 expression in the group of mahuangfuzixixin is lowest(p<0.01), GATA-3 expression in the group of mahuangfuzi,mahuangxixin and dexamethasone are lower than the group of mahuang and model group(p<0.01).Expression of GATA-3 in group of mahuangfuzi,mahuangxixin and dexamethasone are no significant difference(p>0.05).There is no significant difference between the group of mahuang and model group.2.Clinical Observation: Through clinical observation of 30 cases of patients with allergic rhinitis, the cure rate is 26.67%, Significant efficiency rate is 53.33%, the effective rate is 16.67%, total effective rate is 96.70%. After statistical analysis, the difference is significant in 30 cases of patients llergic rhinitis symptoms, signs, syndromes score before and after treatment.Conclusions: 1. Mahuangfuzixixin decoction to some extent can improve the secretion of guinea pig urinary-17 hydroxyl cortisol and serum cortisol,particularly in the use of mahuang and fuzi is most significant.Description mahuang and fuzi to some extent can improve immune function in guinea pigs.2.Mahuangfuzixixin decoction in regulation of Th1/Th2 is by suppressing GATA-3 expression at the transcriptional level, regulation Th0 to normal Th1/Th2 differentiation, thereby suppressing allergic rhinitis Th2 subsets hyperthyroidism, reduce airway inflammation.3.Jiaweimahuangf uzixixin decoction can effectively ease or lift the asthenia cold sysdrome of Allergic rhinitis' nasal itching, sneezing, runny nose, stuffy nose and other symptoms,it is an effective method for treating asthenia cold sysdrome of allergic rhinitis and it deserves further study.
