| Objective:Research in xinjiang uygur and han nationality allergic rhinitis (A11ergicRhinitis, AR) patients mononuclear cells in the transcription factor T-bet and GATA-3expression level, so as to explore the the role of T-bet and GATA-3in AR.Methods:(1)Collection of100patients with allergic rhinitis patients, including uygur and han every50cases),and100healthy volunteers.(2) Separation outside individual nucleus cell (PBMC),with immune magnetic bead method extraction CD4+T lymphocyte.(3) Use SYBRGreen I real-time fluorescent quantitative polymerase chain reaction (PCR) test T-betgenes and GATA-3gene expression level.Results: Allergic rhinitis group GATA-3isobviously higher than that of the control group (P <0.05); T-bet gene mRNA expressionin Allergic rhinitis group is higher than the control group, but there were no significantdifference (P>0.05). Uyghur and Han patients with allergic rhinitis T-betã€GATA-3GenemRNA expression level were no significant difference (P>0.05); Uygur allergic rhinitispatients and healthy people T-betã€GATA-3Gene mRNA expression level were nosignificant difference (P>0.05);T-bet gene mRNA expression level difference was notstatistically significant between Han healthy crowd and patients with allergic rhinitis (P>0.05), But GATA-3gene mRNA expression higher than healthy group (P<0.05).Cnclusions:(1)T-bet〠GATA-3gene SYBR Green I real-time fluorescentquantitative polymerase chain reaction (PCR) were established successfully.(2) Allergicrhinitis patients exist in peripheral blood cells Th2relevant transcription factors GATA-3high expression. T-bet mRNAand GATA-3mRNA have imbalanced expression,and thismay be the important mechanism of allergic rhinitis. Re-establish this balance mayprovide new perspectives for systemic treatment of allergic rhinitis.... |
