Objective:To observe the difference of TET2 and MEG3 gene expression and methylation conditions of MEG3 in aute myeloid leukemia(AML) and normal people, explore the connection in two genes and the clinical significance of the TET2 and MEG3 m RNA expression levels and methylation conditions of MEG3 in AML.Methods:Samples of bone marrow from 60 primary AML patients in People's Hospital of Hainan Province and 10 healthy controls were collected. Measuring expression level of TET2 and MEG3 m RNA in the samples, we assessed TET2 and MEG3 expression change between patients and the normal people and found the relation of the two genes. We also use the matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS) analysis to detect the methylation conditions of the samples, and we analyzed the distribution of the Cp G units among the cases and controls. The distribution of the Cp G units of MEG3 gene associton with the TET2 gene was analyzed, too.Results:(1)TET2 gene expression level of AML patients was lower than the healthy control(p<0.05). MEG3 m RNA level at AML patients was lower than that at the control(p<0.05). The methylation of MEG3 gene in AML patients is higher than the control.TET2 gene m RNA expression have positive correlation with MEG3 m RNA expression, and MEG3 promoter methylation degree have significant negative correlation with TET2 m RNA expression.Conclution:(1)TET2 gene expression level of AML patients was lower than the healthy control,indicate that TET2 gene expression level may be involved in the pathogenesis of AML.(2)MEG3 gene expression level of AML patients was lower than the healthy control, indicate that MEG3 gene expression level may be involved in the pathogenesis of AML.(3)The methylation of MEG3 gene in acute myeloid leukemia is higher than normal people, indicate that mthylation of MEG3 gene may be involved in the pathogenesis of AML.(4)The high methylation of MEG3 gene may have relations with low gene expression of TET2 gene, indicate that TET2 gene can regulate the expression of MEG3 gene.This is need to further validation.
