Effect Of Gacexate Mesilate And Large Dose Of Ambroxol On LPS-Induced Acute Lung Injury In Rats

Objective:To investigate effect of a synthetic protease inhibitor gabexate mesilate and large dose of ambroxol on LPS-induced acute lung injury in rats. Methods:Adult male sprague-Dawley rats(300-350 g) were randomly divided into the nomal control group(C group), lipopolysaccharide group(LPS group), ambroxol hydrochloride group(AMB group, 20 mg/Kg, iv), gabexate mesilate group(GM group, 10 mg/Kg, iv) and ambroxol hydrochloride + gabexate mesilate group(GM and AMB group). The acute lung injury(ALI) model of rats was established by intraperitoneal injection of LPS(5 mg/kg). The general state of rats was observated. The rats were sacrificed 4 h after the injection of LPS. The lungs were took out. The morphology of pathological sections was observated under microscope. The levels of TNF-?, IL-1? and IL-6 in the homogenate of lung were test by ELISA assay. Results:After the injection of LPS, the rats the following symptoms were appealed in the rats, including cyanosis lips, deep and rapid breath, horripilation, reduced acticities, loose stools and no strength to resist or escape. Compared with the LPS group, the symptoms were relieved, the levels of inflammatory factors and the ratio of lung wet dry were also decreased after the injection of gabexate mesilate or large dose of ambroxol( P?0.05). Compared with the AMB or GM group, the symptoms were relieved, the levels of inflammatory factors and the ratio of lung wet dry were also decreased in AMB + GM group( P>0.05). Conclusion:Gabexate mesilate or large dose of ambroxol hydrochloride protects the LPS-induced acute lung injury in rats. There is a obvious synergistic effect of gabexate mesilate combination with large dose of ambroxol on LPS-induced acute lung injury in rats.
