Objective The aim is to establish the the nude mice ectopic intraperitoneal metastatic model with human gastric cancer cell lines MGC-803, and to detect the expression of caveolin-1 and HIF-1? in the tumors and internal organizations so as to provide some experimental evidences for the diagnosis of gastric cancer and detection of distant metastatases.Methods Human gastric cancer cell lines MGC803 were cultured in RPMI 1640. Each BALB/C nude mice were implanted MGC-803 cells by injection and to observe the tumor formation regularly.BALB/C(nu/nu) nude mice were randomly divided into two groups:Control group(10 Injected with normal saline 0.2m L each mice) and treated group(25 Injected with MGC-803 cells 0.2m L each mice).The speciments were fembedded in paraffin and sections were cut by routine procedures for stained with hematoxylin and eosin(H&E). The expression of Caveolin-1 and HIF-1? protein and m RNA were detected by IHC and RT-PCR.Results The nude mice ectopic intraperitoneal metastasis model with human gastric cancer cell lines MGC-803 was established successfully.The IHC experiments showed that the positive rates of caveolin-1 in noncancerous mucosa and gastric cancer were 80%(8/10), 35%(6/23),in normal liver and metastatic liver were 60%(6/10),28%(5/23),in normal lung and metastatic lung were 70%(7/10),40%(3/15),which have significant difference(P<0.05).The IHC experiments showed that the positive rates of HIF-1? in noncancerous mucosa and gastric cancer were 10%(1/10),78%(18/23),in normal liver and metastatic liver were 30%(3/10),65%(15/23),in normal lung and metastatic lung were 20%(2/10),67%(9/15),which have significant difference(P<0.05).RT-PCR experiments showed that the expression of caveolin-1 m RNA in noncancerous mucosa, normal liver and normal lung were lower than those in gastric cancer, metastatic liver and metastatic lung respectively. And the expression of HIF-1? m RNA in noncancerous mucosa, normal liver and normal lung were higher than those in gastric cancer, metastatic liver and metastatic lung respectively. There was negative correlation between HIF-1?and caveolin-1 expression in gastric cancer(r=-0.392,P<0.05).Conclusion(1)The establishment of the nude model with human gastric cancer cell line MGC-823 shows the characteristic of easy operation and high transfer rate, which provides another good stage for the study of the metastasis treatment mechanism.(2)The expression of Caveolin-1 and HIF-1? gene were the important factors in the gastric cancer may be a new biomarker for the evaluation of malignant degrees and liver and lung metastasis of gastric cancer.
