| Objective:to provide an ideal animal model for research in the field of lymph nude metastasis in nude mice and to screen human gastric cancer cell line with spontaneous highly metastasis.Method:the human gastric carcinoma cell line SGC-7901was inoculated into claw pad of nude mice model for establishing lymph node metastasis. The lymph nodes from popliteal fossa, inguinal groove collected at5weeks after tumor implantation were used to observe the tumor formation rate, growth and metastasis by HE staining and immunohistochemistry of CD44v6, EpCAM. Then SGC-L, which was highly metastasis, was obtained by cultured in vitro from the Metastatic lymph nodes. PCR, Western Blot, Scratch Assay, Invasion Assay were employed to compare investigate the behavior of SGC-L and SGC-7901.Result:The tumor formation rate was rate. After5weeks, we found metastasic lymph nodes in popliteal fossa, inguinal groove by HE. In immunohistochemistry, CD44v6, EpCAM, HER2were strong positive. Then we screened successfully a new gastric cancer cell line, which named SGC-L, from the metastasic lymph nodes. In vitro test, the SGC-L have significantly higher ability of migration, invasiveness, metastasis than SGC-7901.Conclusion:The nude mouse model of lymphatic metastasis is successfully established through injecting human gastric cancer cell line to the claw pad. This well characterized line and its vitro expermental should useful to vestigate the mechanisms of lymph node metastasis and to provide a new therapeutic approch for human gastric carcinoma. |
