ObjectiveTo detect the expression of h TGF?1 gene in Hamstring tendon mediated by adenovirus after rabbit's ACL reconstructionMethodsAd-h TGF?1 and Ad-GFP were diluted to 5×108 PFU/m L with DMEM. Forty-eight New Zealand white rabbits were divided into three groups randomly for ACL reconstruction with hamstring tendon autograft. Hamstring tendon was cultured and transfected with Ad-h TGF?1(group A) and Ad-GFP(group B) for 12 hours before ACL reconstruction, and was cultured with DMEM in group C. After 12 hours of transfection, the expression of green fluorescence was observed in groups A and B under fluorescence microscopy. h TGF?1 protein level was detected by ELISA in group A. The rabbits of each group were killed at 2, 4, 8, 12 weeks after operation, and the Hamstring tendon was evaluated by q RT-PCR and Western blot.ResultsGreen fluorescence was observed after 12 hours of transfection in groups A and B. h TGF?1 protein level reached(221.0±12.2) ng/m L at 12 hours in group AThe observation of Anatomy:The rabbits were killed at 2 weeks, 4 weeks, 6 weeks and 8 weeks after operation. There were no signs of fever, swelling and wound infection around the knee. In the article, the synovial significant proliferated and the meniscus were intact. The tendon graft after ACL reconstruction has good tension and fixation. At 2 weeks, there is still visible gap between the tendon graft and the bone tunnel. At 4 weeks, scar tissue could be observed in the inner mouth of the bone tunnel. At 6 weeks, scar tissue in the inner mouth of the bone tunnels was more than before. At 8 weeks, bone tissue could be observed in the external mouth of the bone tunnel.q RT-PCR:h TGF?1 gene m RNA expression could be detected with q RT-PCR methods in group A. However, it could not be detected in group B and group C.Western blot:The expression of TGF?1 protein of group A was higher than the group B and group C, and there was a significant statistical difference(P <0.05).ConclusionAd-h TGF?1 can transfect the Hamstring tendon successfully, and it can effectively express after the rabbit's ACL reconstruction.
