On The Protective Effect To The Intervertebral Disc Degeneration By The Ad/CMV-hTGF-β1 Gene In The Rabbit Body | Posted on:2006-08-23 | Degree:Master | Type:Thesis | Country:China | Candidate:M Wu | Full Text:PDF | GTID:2144360152498968 | Subject:Human Anatomy and Embryology | Abstract/Summary: | PDF Full Text Request | Objective:The research on the protective effects to the rabbit intervertebral disc degeneration by Ad/CMV -hTGF-β1 gene is chiefly for the study of the possible effects to the collagen I ,collagen II and FGF genes in the marrow cells of intervertebral disc by the Ad/CMV-hTGF-β1,and raise its possible functioning mechanism,offering a theoretical evidence in the biological prevention and treatment of intervertebral disc degeneration.Method:20 pure adult New Zealand rabbits are randomly divided into two groups with lOrabbits in each (One group is for the experiment and one for the contrast.).The operation is carried out from the abdomen to expose the waist intervertebral discs,and then 20 micro litter of experimental Ad/CMV-hTGF-β1(with 6×10~6 pfu) is directly injected into the mucleus pulposus between No.4 and No.5 intervertebral discs. The rabbits of the contrast group just accept normal operations without having their interverteebral discs treated. 3 weeds after the operations, the intervertebral discs are taken out and have them sliced to observe the cytological modality of the nucleus pulposus tissue through HE pigmentation. Using the chemical modality of the tissue-immunity to reveal the changes of collagen I, collagen II and FGF in the nucleus pulposus of the intervertebral discs and then VIDAS system is used conduct the semi-ration analysis.ResultrThe experimental results through HE pigmentation show no difference in the state of intervertebral disc nucleus pulposus. The experiment through the chemical modality of the tissue-immunity shows that the expression of collagen I of the nucleus pulposus in the injecting hTGF-β1 gene group is lower than that in the contrast group (The group for the experiment: OPTDM=0.1804±0.0211; The group for contrast: OPTDM=0.2768±0.0161; P<0.05),but the expression of collagen II and FGF protein in the nucleus pulposus is higher than that in contrast group (The group for the experiment: OPTDM=0.6936±0.0312; The group for contrast: OPTDM=0.5068±0.0235: P<0.05).
