Objective: To investigate the effects of mi R-711 on the invasion, metastasis and epithelial mesenchymal transition(EMT) of human gastric cancer cells MGC-803 and SGC-7901.Methods: MiR-711 mimics, mi R-711 inhibitors and mi RNA empty plasmids were transfected into gastric cancer cell MGC-803 and SGC-7901 by liposomes transient transfection method, MiRNA empty plasmids transfection group was taken as the negative control group, the non-transfection group taken as blank control group. After 48 h, the effectiveness of transfection was observed by under fluorescence microscope. Cell scratch experiment and Transwell invasion experiment were used to detect the migration and invasion. Western blot was used to observe the expression of epithelial protein and mesenchymal protein of transfected MGC-803 cells and SGC-7901 cells.Results:1.The transfection efficiency of MGC?803 cells and SGC?7901 cells observed under fluorescence microscope was more than 75%.2.As revealed by Transwell invasion test of MGC-803 cells:the number of invaded cells in NC and K groups were(120.00±4.58) and(119.67±5.13), while that was significantly lower of the mi R-711 mimics transfection group( 70.67 ±5.51),and was significantly higher of the mi R-711 inhibitors transfection group(213.67 ±9.07),( P<0.05); As revealed by Transwell invasion test of SGC-7901cells:the number of invaded cells in NC and K groups were(114.67 ±7.37)and(109.67 ±5.51), while that was significantly lower of the mi R-711 mimics transfection group(63.66 ±5.51),and was significantly higher of the mi R-711 inhibitors transfection group(188.33 ±8.50),( P<0.05).3.As revealed by the cell scratch test of MGC-803 cells: at 48 h after scratch, the healing rate of NC and K groups were(32.52±1.73)% and(32.15 ±1.55)%,while that was significantly lower of the mi R-711 mimics transfection group(7.08±0.73)%, and was significantly higher of the mi R-711 inhibitors transfection group(49.03±1.59),( P<0.05); As revealed by the cell scratch test of SGC-7901 cells: at 48 h after scratch, the healing rate of NC and K groups were(28.74±1.45)% and(28.22 ±2.49)%, while that was significantly lower of the mi R-711 mimics transfection group(4.76 ±0.40)%, and was significantly higher of the mi R-711 inhibitors transfection group(45.79±0.23),( P<0.05).4.As revealed by Western Blot test of MGC-803 cells and SGC-7901 cells :compared with NC and K groups, the relative expression of E-cadherin protein was increased in the mi R-711 mimics transfection group, while the expression of Vimentin protein was decreased,( P<0.05). Compared with NC and K groups, the relative expression of E-cadherin protein was decreased in the mi R-711 inhibitors transfection group,while the expression of Vimentin protein was increased,(P<0.05).Conclusion: MiR-711 overexpression could inhibit the invasion, metastasis and EMT of human gastric cancer cells.
