ObjectiveTo measure the expressions of Survivin and Klotho of cancer tissue and adjacent tissue in the gastrointestinal neuroendocrine carcinoma patients and explore the relationship between expressions of Survivin and Klotho and prognosis of gastrointestinal neuroendocrine carcinoma. Methods51 cases data of gastrointestinal neuroendocrine carcinoma patients were retrospectively studied. Expressions of Survivin and Klotho of cancer tissue and adjacent tissue in the gastrointestinal neuroendocrine carcinoma patients were measured by immuno histochemistry and Western blot. The relationship between expressions of Survivin and Klotho and prognosis of gastrointestinal neuroendocrine carcinoma was analysed. Results1. In 51 cases gastrointestinal neuroendocrine carcinoma patients, Survivin expression was detected in cancer tissue in 35 cases patients and expression rate of Survivin was 68.63%. Expression of Survivin was not found in the cancer adjacent tissue in all patients. The expression level and expression rate of Survivin in gastrointestinal neuroendocrine carcinoma patients with small cell carcinoma were significantly increased compared with the large cell carcinoma patients. Expression of Survivin was related with tumor stage and tumor differentiation, but there were no relationship between expression of Survivin and other factors such as age, gender, etc. Expression of Survivin in poorly differentiated and undifferentiated cancer tissues was significantly higher than that in highly differentiated cancer(P=0.0013). Expression rate of Survivin was 100% in clinical stage III(4/7) and stage IV(12/12 patients and was significantly higher than that in clinical stage II(15/27) and stage I(1/5) patients(P=0.0023).2. In 51 cases gastrointestinal neuroendocrine carcinoma patients, Klotho expression was showed a weak positive reaction in cancer tissue in 21 cases patients by immuno histochemistry. Klotho expression was showed a strong positive reaction in cancer adjacent tissue in all patients by immuno histochemistry. Klotho expression measured by Western blot was significantly decreased in cancer tissue of gastrointestinal neuroendocrine carcinoma compared with the cancer adjacent tissue. Expression of Klotho was related with tumor stage and tumor differentiation, but there were no relationship between expression of Klotho and other factors such as age, etc. Expression of Klotho in poorly differentiated(0/6) and undifferentiated(0/4) cancer tissues was significantly lower than that in highly differentiated(6/7) cancer(P=0.0035). Expression rate of Survivin was 100% in clinical stage I(5/5) patients and was significantly higher than that in clinical stage II(13/27) and stage III(2/7) patients(P=0.0067). Expression of Klotho was not detected in stage IV patients. Conclusions1. In gastrointestinal neuroendocrine carcinoma patients, expression of Survivin in cancer tissues in small cell carcinoma is significantly increased compared with the large cell carcinoma. Expression of Survivin in cancer tissues in large cell carcinoma is significantly increased compared with the cancer adjacent tissue.2. In gastrointestinal neuroendocrine carcinoma patients, expression of Klotho in cancer tissues in small cell carcinoma is significantly decreased compared with the large cell carcinoma. Expression of Klotho in cancer tissues in large cell carcinoma is significantly decreased compared with the cancer adjacent tissue.3. Expressions of Survivin and Klotho are related with tumor stage and tumor differentiation. Prognosis is poor in the patients with Survivin positive expression and Klotho negative expression Survivin and Klotho are important indicators for predicting and evaluating the prognosis of patients with gastrointestinal neuroendocrine carcinoma.
