Objective:To study the effect of SiJunZi Decoction on defecation status and serum MT level of STC rats, and to provide the laboratory basis for the treatment of STC with SiJunZi Decoction.To study the regulation mechanism of melatonin in the pathogenesis of STC, and provide a new treatment method for clinical treatment of STC.Methods:A total of 60 healthy male SD rats were randomly divided into 4 groups,namely normal group, model group, mosapride group and SiJunZi Decoction group, 15 rats in each group. In addition to the normal group, the rats were using compound diphenoxylate(8mg/kg·d) induced STC model, modeling time 120 days.After successful modeling, the use of corresponding drugs or distilled water to intervene. After the intervention, the general situation of each group,the frequency of defecation,and the weight of wet stool were observed.The serum MT levels of rats in each group were detected by ELISA.Results :(1)Compared with the normal group,the weight of model group was significantly reduced(P?0.05),the defecation frequency of model group was significantly reduced(P? 0.05),the weight of wet stool in model group was significantly reduced( P ? 0.05),the serum level of MT in model group was increased significantly( P ? 0.05).(2)After treatment,the weight of rats in Sijunzi Decoction group were obviously higher than those in the model group(P?0.05), and the effect is better than mosapride group.(3)Compared with the model group, after treatment,in SiJunZi decoction group and mosapride group, the number of bowel increased significantly(P?0.05), the wet weight of fecal pellets increased significantly(P?0.05),and the SiJunZi decoction group increased the effect of fecal water content was better than mosapride group( P? 0.05).(4)Compared with model group,after treatment,the serum levels of MT in SiJunZi Decoction group decreased significantly(P?0.05),reaching with the normal group and better than that in mosapride group(P?0.05).Conclusion:The incidence of STC may be related to the abnormal increase of serum MT levels. SiJunZi Decoction can significantly improve the symptoms of constipation in STC rats, shorten the discharge time of intestinal contents.The mechanism of action maybe to regulate the abnormal elevation of serum MT levels...
