Objective: To investigate whether the 1-methylhydantoine can inhibit the proliferation of Breast cancer MCF-7 and MDA-MB-231 cell line, and to explore the possible ways to inhibit the proliferation of breast cancer cells with 1-methylhydantoine.Methods: Taking breast cancer MCF-7 cell line and MDA-MB231 cell line cultured in vitro as the general researching object, use the MTT method to detect the effect of the growth status of two kinds of breast cancer cells which are intervened by 1-methylhydantoine and octreotide, and the invasive ability of them by transwell assay for drug determination. To detect the expression of CAMP, Bcl-2 and Bax protein in cells after Western-blot drug action. Use the immunohistochemistry method to detect the effect of 1-methylhydantoine on the expression of MCF-7 cells and MDA-MB231 cells SSTR2.Results: 1. 1-methylhydantoine can inhibit the growth of breast cancer cells and the invasion ability. The differences were statistically significant(P < 0.05).2. 1-methylhydantoine can inhibit the expression of cell c AMP and Bcl-2 protein, and promote the expression of Bax protein(p<0.05).3. SSTR2 was expressed in two breast cancer cell lines. Under the action of 1-methylhydantoine, the expression of SSTR2 was higher than that of the control group, and there was a trend of increase with the increase of the drug concentration.Conclusion: 1. 1-methylhydantoine can inhibit the proliferation of breast cancer MCF-7 and MBA-MB-231 cells, and it also can inhibit the invasion of them.2. It will Produce complex biological effects in cells after the action of 1-methylhydantoine, maybe by affecting the expression of c AMP, Bax, Bcl-2 protein in cells, activate of SSTR in the cell and play a role in inhibiting the proliferation of breast cancer cells.3.
