Bacterial infections can occur in different parts of the body,is a common disease clinical departments,as people non-standard use of antibiotics and the abuse of antibiotics,bacterial drug resistance phenomenon more and more.Eliminate the infection and the spread of the infection control is very important.Most people believe that,Chinese herbal medicine safe,efficient,no drug resistance,in recent years,some clinical doctors found that traditional Chinese medicine(TCM)in the mid-late"anti-infection treatment,curative effect is reduced.The researchers studied,found that a variety of bacteria existing in single Chinese herbal medicine and Chinese herbal medicine formula resistance phenomenon.At present,many scholars have focused on the study of the bacteria resistant to antibiotics,the study found that the bacteria are resistant to antibiotics mainly biofilm formation,the formation of biofilms,drug resistance bacteria through many ways to prevent antibiotic,gene mutation and gene recombinant bacteria itself,outside system such as antibiotics.Studies have confirmed that the bacteria on the traditional Chinese herbal medicine drug resistance,but the mechanism of drug resistance is still not clear,so the experiment research.Objective: explore Chinese medicine drug-resistant e.coli biofilm formation ability and the transmission of resistant plasmids and regulation.Methods: extracted from the standard urine of patients with clinical pathogenic bacteria,and identification,determination of identified as experimental strains of drug-resistant bacteria Wen YangTong pour soup for minimal inhibitory concentration(MIC),the experimental strains in 1/2 MIC experimental drug M-H broth medium amplification,switched to a cultivation every 3 days,9 days to pick a single colony to 4times the MIC medicated tablet,determine its resistance.1/2 MIC induce drug-resistant bacteria,4MIC screening,extracting plasmid drug-resistant bacteria;Turn resistance plasmid to Trans5 ?,coating on the 4MIC medicated tablet,to observe the growth ofcolonies;Will Trans5 ?,Trans10 are coated in 6.2 g/L medicated tablet,observation colony growth;Turn resistance plasmid to Trans10,coating on the 4MIC medicated tablet,to observe the growth of colonies;Pick 6 two monoclonal strain 5028,4996,1/2MIC induction,4MIC screening,extracting its plasmid,bacterial plasmid extraction transformation at the same time,the observation plasmid changes;With drug resistant strains and sensitive strains group,according to the literature method,the experimental strains for LB culture medium,the formation of biofilms,flush with NaCl solution,1%crystal violet stain,distilled water rinse,dry,acetic acid dissolves,measured at the wavelength of 570 nm absorbance of average;In traditional Chinese medicine drug resistance and sensitive group,analysis of drug resistant strains and sensitive strains biofilm formation ability strong and the weak.Results: drug resistant strains of WenYangTong pour soup minimal inhibitory concentration(MIC),50 g/L;Of 3 strains induced by screening e.coli plasmid extraction,transformation,the result is shown in figure 2,the resistant plasmids into Trans5 ?,after training on the 4MIC tablet,bacteria can grow in the medicated tablet on normal;Trans5 ?,Trans10 are coated in 6.2 g/L medicated tablet,all can normal growth on the plate;Resistant plasmid into Trans10 after training on the 4MIC tablet,after conversion bacteria can grow on the medicated tablet normal;Into the bacterial plasmid extraction,and monoclonal 5028,4996 plasmid is equal;Bar charts found by biofilm formation ability,form a biofilm ability resistant group was obviously stronger than the sensitive group,biofilm morphology showed resistant biofilm area significantly greater than the sensitive group.Conclusion: e.coli 1351 5028 4996 WenYangTong drench Chinese medicine drug resistance;Resistance gene is located in a plasmid,plasmid can steady transfer between different strains,strains resistant to obtain resistant plasmid;Biological membrane mediated drug-resistant bacteria.
