Echinacea(Echinacea Moenck)is a perennial asteraceae herbaceous wild flowers,dicotyledoneae.It has the function of regulating immunity,anti-inflammatory,antioxidant,antibacterial,antiviral and anticancer.It was known as the world's"immune" herbsthe,It was first used by American Indians,for a variety of infections,tuberculosis,insect and snake bites.Now Echinacea is widely used for immune regulator and promoter.Endotoxin is Escherichia coli gram-negative cell wall components,lipopolysaccharide(LPS).It is released by the lysis of bacterial death.when a large number of bacterial died,large amounts of endotoxin is release into the blood,resulting in endotoxemia,systemic excessive inflammatory reaction,the body's cells,tissues and organs injury,and even causing the death of the body.So harm to human.In this study,the effect of Echinacea purpurea polysaccharide on intestinal epithelial cell line IEC-6 after LPS injury in vitro was studyed.As to make clear Echinacea polysaccharide on the mechanism of IEC-6 cells after stimulated by LPS.Some methods were used to detect a few of index,MTT assay for the rate of cell proliferation of IEC-6 cells;RT-PCR method for the expression leve of IEC-6 cells secrete cytokines TNF,IL-1,IL-6 and IL-8 mRNA;Western blot assay for the protein expression level of IEC-6 cells secrete cytokines NF-?B p65,pI?B?;In vivo,LPS was used to induce mouse model of endotoxin damage and inflammation to study the effets of Echinacea purpurea polysaccharide on endotoxin damage.The expression levels of serum cytokines TNF-?,IL-1?,IL-6 were detected by ELISA method.The paraffin sections of lung tissue for the severity and dry and wet weight for the severity of pulmonary edema were tested;The real-time quantitative PCR method detected COX-2?iNOS?HO-1.NF-?B mRNA expression levels of lung tissue.In this animal models,the mechanism of Echinacea purpurea polysaccharide on endotoxin damage was studied,so as to make sure whether Echinacea polysaccharide can provide a protective effect on the endotoxin injured mice.The results showed that the proliferation of IEC-6 cells can be enhanced by the pretreatment of Echinacea polysaccharide after LPS damage,The expression level of TNF-?,IL-1?,LL-6,IL-8 mRNA and NF-?B p65,pI?B? of IEC-6 cells secretion after LPS injury be inhibited by Echinacea polysaccharide.Echinacea polysaccharide reduced the damage of IEC-6 cells,and has protective effect on the cells..the levels of inflammatory factors TNF-?,IL-1?,IL-6 in Serum and COX-2,iNOS,HO-1,NF-?B mRNA in the lung tissues were effectively inhibited by Echinacea polysaccharide.
