| Objective: Depression is a form of attack of manic depression, which is one of the most common mental illnesses, and the incidence of depression have a significant differences in gender, especially in the menopausal transition the estrogen sharply reduces, the incidence of female depression significantly higher than the male.in recent years, people find that exercise without any side effects as one of interventions can alleviate this phenomenon, but the inner mechanism of movement of antidepressant is unclear. Therefore, in this subject, I would build an ovarian mice model(OVX mice model) to imitate the status of estrogen lacked,then giving treadmill exercise intervention, using the classic way which is to detect the depression like behavior to tests the change in depression in mice.Using molecular biology experiment method, testing the index changes of inflammation in OVX mouse brain,at the same time analysising the correlation between the indicators change and the body’s estrogen level, further explore the Mechanism of exercise Improve depression like behavior caused by a lack of internal estrogen in mice’s activity, and enrich the depression theory and related aspects of knowledge, which also has important significance to promote China’s national sports of fitness plan.Design: 60 healthy female C57 / BL6 mice can be divided into four groups: control group(Sham),Sham and exercise group(Sham-Ex), ovarian group(OVX), ovarian and exercise group(OVX-Ex), according to the situation of body weight, blood corticosterone levels and uterus weight to determine whether mice to ovarian surgery success.To postoperative recovery for a week,Con group and OVX group without any training, Sham-Ex group and OVX-Ex group begin to 5 weeks to treadmill exercise,exercise plan is as follows:The first day of the first week to given adaptability treadmill exercise(15m/min, 20min), the speed is change but continuous increaseing10 min every day from the second day,the fifth day 5 given adaptability treadmill exercise(15m/min, 40min), then rest two days. From 2 weeks to 5 weeks given formally running exercise(15m/min, 45min/d, 5d/week);at the end of the exercise intervention, using sucrose preference test, open field test, O maze test, forced swimming test and tail suspension test to test depression like behavioral in four groups mice, Behavioral test is over, put to death animals, collect serum and hippocampus, using Elisa, Real-time PCR and Western blot to test the index changes of inflammasome in mice.Results: 1、compared with the Sham group, the weight of OVX(p<0.01) obviously increase, the weight of the uterus(p<0.01) and serum estradiol(E2) levels(p<0.01)significantly reduced, and given the OVX mice exercise intervention can reverse the above phenomenon; Compared with OVX group, the OVX-Ex(p<0.01) significantly reduced weight, serum estradiol(E2) levels(p<0.01) significantly increased.2、Compared with the Sham group, Sham-Ex are similar(p>0.05),the OVX of 24 h sucrose preference(p<0.01), the immobility time(p<0.01)in O-MAZE and the center distance(p<0.01)and number of crossing(p<0.01) in OFT significantly decreased,there was a increased in the immobility time(p<0.01)in TST and the floating immobility time in forced swimming.given the OVX mice exercise intervention can reverse the above phenomenon; Compared with OVX group, the OVX-Ex of the 24 h sucrose preference(p<0.01),the immobility time(p<0.01)in O-MAZE and the center distance(p<0.01)and number of crossing(p<0.01) in OFT significantly increased, there wasa decreased in the immobility time(p<0.05)in TST and the floating immobility time in forced swimming.3、The m RNA level and the protein expression level of of inflammatory factor s within the hippocampus changes: compared with the Sham group, the m RNA level and the protein expression level of NLRP3, Caspase-1, IL-1β, IL-18 in Sham-Ex has no obvious difference(p>0.05), the OVX was significantly increas ed(p<0.05 or p<0.01),compared with OVX group, the OVX-Ex decreased signif icantly(p<0.05).Conclusions:1) OVX can lead to weight gain in mice, reducing the content of serum estradiol(E2),and increasing depression like behavior in mice.At the same time,which can increase the m RNA levels and the protein levels expression of inflammatory factors such as NLRP3, ASC, Caspase-1, IL-1β, IL-18.2)Treadmill exercise can reverse the weight in OVX, increasing the content of serum estradiol(E2), alleviate depression behavior in mice. At the same time by cutting the m RNA levels and the protein expression of inflammasome factors such as NLRP3,Caspase-1, IL-1β, IL-18 and then decreasing the protein levels of them. |
