Objective:To study the effect of smokeless tobacco extracts(ST)on adhesion and proliferation of human gingival fibroblasts(HGFs)cultured on pure titanium surface,to lay a theoretical basis for how to improve planting rate of smoking patients clinically.Method:1.The fatal HGFs were cultured with reformed enzymatic digeston.2.HGFs were identified by immunohistochemical SP method,and drawing the growth curve.3.Using metallographic sandpaper grind the titanium plate,and observing its microstructure with metallographic microscope after measuring surface roughness.4.Detection changes of the adhesive rate of HGFs by CCK-8 on pure titanium surface after intervention with ST.5.Detection effect of ST on the Long-term proliferation of HGFs by CCK-8 on pure titanium surface.6.Detection effect of ST on the production FN of HGFs by ELISA on pure titanium surface.Result:1.The HGFs cultured in long and stringy,which accords with the cell growth in vitro.SP method shows that vimentin expressed positive and cytokeratin expressed negative in HGFs.2.The parameter of Ra of the titanium Plates is: Ra =(0.187 ± 0.031)?m,the surface of titanium plate was polished uniform microscopically,which is suitable for cell growth.3.CCK-8 shows that adhesive rate of HGFs is on the decline with elevated concentration of ST,the inhibition is of significance when ST concentration is higher than 5.0 g/L.(p<0.01)4.CCK-8 shows that ST inhibits the proliferation of HGFs on pure titanium surface in time and concentration dependence.5.ELISA shows that the inhibition effect of ST on FN is enhanced gradually with the increase of concentration and time,and the inhibition in 2.5g/L is strongest,the effect is longest.(p<0.01)Conclusion:1.HGFs cultured in vitro successfully,and the cells grow well and stable.2.ST can inhibit adhesive and proliferation of HGFs on pure titanium surface,and the mechanism may be related to the decrease of FN secreted by HGFs.
