Background and Objective Viral myocarditis is an inflammatory disease,in which the heart was infected by virus. However the pathogenesis of VMC is not clarified thoroughly, the therapy needs further improvement. This study is to establish the modals of VMC through Balb/c mice infected with coxsackievirus B3, make andrographis injection as treatment intervention, then observe the protective effect and discuss the mechanism of andrographolide on viral myocarditis in mice modelMethods 1. A total of 150 male Balb/c mice were randomly divided into three groups :normal control group(n=40), viral myocarditis model group(n=60) and andrographolidetreated group( n = 50). 2. Viral myocarditis model group and andrographolide-treated group were intraperitoneally inoculated with CVB3 to induce myocarditis,control group with virus-free Eagle' s medium instead.Andrographolide-treated group was given andrographis injection through intraperitoneal injection everyday from 0.5 hoursafter injecting virus for 14 days,control group and myocarditis group were used normal saline instead. 3. The general state of the mice were observed, deaths were recorded and calculated the survival rate of mice; the levels of TNF-? in myocardial tissue and serum IL-6 were assayed by immunohistochemical stain and ELISA respectively.Results 1. At 7 and 14 days after CVB3 infection,compared with the control group, serum IL-6 level ?expression of TNF-?and the pathological integral were increased in viral myocarditis model group( P < 0. 05). 2. At 7 and 14 days after CVB3 infection,compared with the model group, serum IL-6 level ?expression of TNF-?and the pathological integral were decreased in the andrographolide-treated group( P < 0. 05).Conclusions Andrographolide could reduce the myocardial necrosis in VMC mice infected by CVB3, this might be because of its decrease serum IL-6 level and the expression of TNF-? in myocardial tissue.
