Background and PurposeEsophageal cancinoma(EC) is a common malignant tumor of digestive tract entity, in all the malignant tumors accounted for 2%, ranking seventh in the world, is one of the six most common cancers in our country.Esophageal adenocarcinoma(EAC) is one of the most common pathological type of esophageal cancer with EAC, mainly in western countries, the incidence of EAC in China after the rate of esophageal squamous cell carcinoma(ESCC).Patients with adenocarcinoma of the survival rate is 10%, the postoperative mortality range around the world each year for the 15%-40%. Died of esophageal cancer of about 30 million, but accounted for about 15 million in China. According to the forecast, cancer mortality continues to grow, to 2030 estimated 1.1 million people will die from cancer.At present, including China in the whole world, the rate had an obvious increasing trend occurred in EC. Although studies have shown that early EC underwent surgical resection can achieve satisfactory effect, but because the early symptoms are not obvious, the specificity is not high, but the majority of patients in the early onset of eating choking feeling, pain so don't care about the chest, so most of the patients had been diagnosed in advanced stage. For the diagnosis and treatment of(EAC), to explore a new method and thought is very important. At present, there are many methods for the treatment of esophageal carcinoma, the main way is to use the methods of surgery(lesion and digestive tract reconstruction) according to the actual situation, combined with chemotherapy, immune therapy, comprehensive therapy of Chinese medicine. The above methods as the traditional treatment methods, but not for the advanced patients who underwent radical surgery, especially the resistance By operation, quality of life and survival is not improved obviously, appear illness and death can not tolerate the side effects of treatment.Is targeting, so in the case of esophageal cancer, seeking to explore the development of a new, effective treatment method is very necessary. As technology advances, recombinant DNA technology to gradually perfect, tumor immune treatment technology in the rapid development in the future is likely to become the main methods for diagnosis and treatment of esophageal cancer is a common digestive system malignant tumor of the incidence of multi-stage, multi factor and multi gene interaction results, to study the pathogenesis of esophageal cancer diagnosis and treatment is of great significance. The research for the CA IX in esophageal adenocarcinoma the expression study of esophageal adenocarcinoma incidence mechanism for targeted therapy provides research foundation.Carbonic anhydras IX(CA IX) is a transmembrane protein for tumor associated protein, mainly in normal human digestive tract and pancreas, gallbladder, liver and other organs related expression in other normal tissues almost no expression, but in many solid tumors in highly expressed and involved in tumor cell the proliferation, invasion and metastasis related.CA IX resistant to radiotherapy and chemotherapy, and to accelerate the deterioration of cancer cell proliferation and metastasis. Oxidative phosphorylation rapid growth of tumor cells to anaerobic glycolysis, the tumor microenvironment to ischemia, hypoxia.CA IX tumor cells in alkali acid, the tumor growth and to create the conditions for metastasis and invasion of.CA IX protein in domestic Research for adenocarcinoma is poorly understood, and the current situation there is no domestic report about expression of CA IX in esophageal carcinoma.Therefore, the expression of CA IX in esophageal adenocarcinoma, explore the pathogenesis of esophageal cancer, may provide a new theoretical basis for the clinical diagnosis and prognosis. Objects and MethodsObjective To study the expression of CA IX in esophageal carcinoma and its clinical significance.Methods 1.Methods SP immunohistochemical detection of the expression CA IX proteins in 45 cases of esophageal adenocarcinoma,atypical hyperplasia and corresponding normal tissues; application by RT-PCR analysis of CAIX expression distribution in esophageal adenocarcinoma,atypical hyperplasia and corresponding normal tissues; and separately analyzed the expression level and clinicopathological features relationships CAIX.2.Using statistical software SPSS 17.0 was used for statistical analysis, and the measurement data of normal distribution are expressed by mean plus or minus standard deviation.The two groups were compared using t test, repeated measurement data using repeated measurement data analysis of variance.Count data is used for the frequency and fraction(rate) indicated, ?2 test was used to compare the rates, the correlation of the expression analysis using the Pearson contingency coefficient.Comparison between the two groups using ?2, paired count data using paired ?2 test; P<0.05 difference was statistically significant. ResultsCA IX protein in esophageal adenocarcinoma,atypical hyperplasia and normal tube organization in this next rate dependence 66.7%(30/45),24.4%(11/45),4.4%( 2/45),any two with statistically significant difference between the three(?2=41.845,P<0.05).RT-PCR results showed that CA IX expression in different tissues of esophagus was 0.217±0.017,0.436±0.03,0.544±0.043,two with statistically significant difference between the three(t=60.614 P<0.001). There was relationship in the expression intensity of CA IX protein and the grading of tumor,the depth of invasion,lymph node metastasis,Clinical staging(P<0.05),there was nothing to do with sex and age(P<0.05). ConclusionCA IX protein may be involved in the carcinogenesis and development of esophageal adenocarcinoma,the detection of CA ? is helpful for the diagnosis and treatment of esophageal adenocarcinoma.
