Phyllostachys heterocycla is a traditional Chinese medicine in our country, flavonoids is its main chemical composition. There is research shows that flavonoids from Lophatherum gracile have good anti-inflammatory effect, but the mechanism of anti-inflammatory is not clear. We will rely on the basis of research on anti-inflammatory effects of Phyllostachys heterocycla flavonoids to do in-depth research and discuss the material basis.To optimize the extraction conditions of flavone from Phyllostachys heterocycla. An extraction procedure was optimized by orthogonal test. The content of flavone which was used as the assessment index was determined by UV spectrophotometric method. The optimum extraction process was as follows: To use crude drugs in processed pieces with 10 times 70% Methanol, extraction 3 times and each time lasts 24 h.For purifying the flavonoids from Phyllostachys heterocycla, to use MCI-GEL CHP20 P packing separate the flavonoids extracts, the sample concentration is 0.66g/ml, the adsorption velocity is 1BV/h, 80%methanol solution as the elution soulution, elution velocity is 3BV/h, elution volume is 4BV, finally we get 106 g flavonoids extract powder. For separating the different parts of the flavonoids, to use polyamide packing separate the flavonoids extracts, the adsorption velocity is 1BV/h, 80% methanol solution as the elution soulution, elution velocity is 2BV/h, elution volume is 3-5BV, get in turn, with distilled water, 10% methanol, 30% methanol, 50%methanol, 75% methanol and 100% methanol elution, finally get the distilled water elution part is 34.5 g, 10% methanol elution part is 26.3g, 30% methanol elution part is 7.7 g, 50% methanol elution part is 8.6g, 75% methanol elution part is 5.1g and 100% methanol elution part is 1.1 g.To optimize the the content determination of total flavonoids in different parts of Phyllostachys heterocycla. Using rutin as control, the content of total flavonoids in different parts of Phyllostachys heterocycla. The detection wavelength was set at507 nm. The calibration curve of rutin was linear in the rang of0.01mg-0.20 mg, r=0.9989.The linear equation is Y=17.71X+0.0225.The average recovery was 99.41%, RSD=1.3%(n=6). The method is simple, accurate and reproducible. The content of total flavonoids in different parts from Phyllostachys heterocycla is differences,which is 76.16mg/g,138.35mg/g,467.77mg/g,604.46mg/g,797.26mg/g and 300.95mg/g, provides a scientific foundation for the further study on flavonoids from Phyllostachys heterocycla.To investigate the anti-inflammatory effects of flavonoids from Phyllostachys heterocycla by auricle edema be induced by dimethylbenzene. The dexamethasone(DXM) group, the part 2 to part5 showed significant inhibitory effect on auricle edema induced by dimethylbenzene in mice, compared with the blank control group showed significant difference(P<0.01), the inhibition rate, in order: 61.88, 64.57%, 77.13%, 62.33% and 83.41%, part 1 showed inhibitory effect on auricle edema induced by dimethylbenzene in mice, compared with the blank control group showed difference(P<0.05), the inhibition rate was 43.50%, the part 6 showed uninhibitory effect on auricle edema induced by dimethylbenzene in mice(P > 0.05).To investigate the anti-inflammatory effects of flavonoids from Phyllostachys heterocycla by abdominal cavity capillary permeability be increased by acetic acid. The part 5 showed significant inhibitory effect on abdominal cavity capillary permeability be increased by acetic acid in mice, compared with the blank control group showed significant difference(P<0.01), the inhibition rate was 66.73%, DXM group showed inhibitory effect on abdominal cavity capillary permeability be increased by acetic acid in mice, compared with the blank control group showed difference(P<0.05), the inhibition rate was 53.62%, the others part showed uninhibitory effect on abdominal cavity capillary permeability be increased by acetic acid in mice(P>0.05).To investigate the anti-inflammatory effects of flavonoids from Phyllostachys heterocycla by acute inflammation be induced by air-pouch. The DXM group and part 5 showed significant inhibitory effect on acute inflammation be induced by air-pouch in mice,compared with the blank control group showed significant difference(P<0.01), the inhibition rate was 16.40% and 19.55%, the others part showed uninhibitory effect on abdominal cavity capillary permeability be increased by acetic acid in mice(P>0.05).To investigate the anti-inflammatory effects of flavonoids from Phyllostachys heterocycla by granuloma be induced by cotton pellet.The DXM group and part 5 showed significant inhibitory effect on granuloma be induced by cotton pellet in mice, compared with the blank control group showed significant difference(P<0.01), the inhibition rate was 36.98% and 38.52%, part 4 showed inhibitory effect on granuloma be induced by cotton pellet in mice, compared with the blank control group showed difference(P<0.05), the inhibition rate was 29.12%, the others part showed uninhibitory effect on granuloma be induced by cotton pellet in mice(P>0.05).The major components of part 5 were studied by HPLC/Q-TOF-MS. A Ultimate XB-C18(5?m,3.0×150mm) was used at a flow rate of 1 ml/min.The HPLC gradient was acetonitrile-formic acid(99.9:0.1,v/v)(B)in water-formic acid(99.9:0.1,v/v)(A). HR-MS/MS analysis were carried out using a Micr OTOF-Q II Focus mass spectrometer fitted with an ESI source operating in Auto-MSn mode to obtain fragment ion. TIC chemical profiles of the extract were established in both positive and negative modes. The structures of known components in Phyllostachys nigra were identified by comparing their retention times, molecular weight and CID fragmentation patterns with their corresponding compounds reported in the literature.This experiment optimized the greatest anti-inflammatory effective parts from Phyllostachys heterocycla by animal experiments on the inflammation mice. Through against the material basis for the effective parts of the research, Analyzing the chemical structure of anti-inflammatory activity components in the Phyllostachys heterocycla, which will provide a scientific basis for further research.
