ObjectiveThe aim of this study is to investigate the effects and possible mechanisms of klotho gene in pancreatic cancer.Methods1.The expression of klotho in different cell lines was studied by Western blotting.2.Plasmids encoding klotho or klotho specific siRNA were constructed to overexpress or knockdown klotho gene in Pancl cells.The MTT assays and flow cytometry were used to assess the effects of klotho on cell growth and apoptosis.The activation of Wnt/?-catenin signaling pathway in Panc1 treated by pCMV6-KL and siRNA-kl was measured by Western blotting.Results1.The expression of klotho in normal pancreatic cell lines is higher than pancreatic cancer cells.2.Forced expression of klotho reduced proliferation and promoted the apoptosis of Panc1 cells,whereas klotho silencing in Panc1 cells enhanced proliferation.3.Forced expression of klotho in Panc1 cells inhibited the activation of Wnt/?-catenin signaling pathway.In contrast,the important signaling molecule in Wnt/?-catenin pathway,c-myc and cyclinDl were up-regulated,when klotho was silenced by siRNA-kl.ConclusionOur studies indicate klotho as a potential tumor suppressor in pancreatic cancer,through inhibition of activation of the Wnt/?-catenin pathway.
