Effects Of “Hejiezhitong Granule” On C-fos,c-jun Gene Expression Of Migraine Rat Model

Purpose: Establishment of nitroglycerin migraine rat model,observe the effect of“Hejiezhitong Granule” on the behavior of migraine rat model.Using immunohistochemical staining to observe the effect of “Hejiezhitong Granule” on the expression of c-fos,c-jun in spinal trigeminal nucleus.From the view of gene expression to explore the mechanism of clinical effect of “Hejiezhitong Granule”,and provide evidence for clinical efficacy of "Hejiezhitong Granule" treating migraine.Method: 60 healthy Wistar rats,half male and half female,were randomly divided into 6groups(n=10 in each group): control group,model group,Hejiezhitong Granule group(2.14g/kg/day,1.07g/kg/day,0.54g/kg/day),Flunarizine Hydrochloride Capsules group(0.89mg/kg/day),Flunarizine Hydrochloride Capsules group as a positive control group.The Flunarizine Hydrochloride Capsules and Hejiezhitong Granule were respectively dissolved in Saline by gavage once daily for 15 days,Control and model group received equal volume of Saline solution.Half an hour after the last gavage,each group received subcutaneous injection of nitroglycerin except the control group,and observed the behavior changes of rats.Then sacrificed after 4 hours,collected the brain tissues,prepared for immunohistochemistry.Then the images were captured by a microscope.ImagePro Plus 6.0was applied to analyze the result of c-fos,c-jun.Result: In addition to the control group of rats,the other rats began to scratch head,climb cage,and walk back and forth after subcutaneous injection of nitroglycerin,It means successful model of migraine.The behavioral changes duration in Flunarizine Hydrochloride group and traditional chinese medicine groups were shorter than that in model group(P<0.05).The number of c-fos and c-jun positive cells and optical density of the other five group is higher than that of control group(P<0.05).Compared with the model group,The number of c-fos and c-jun positive cells and optical density of each treatment group is lower than that of the model group(P<0.05).The same indicator of Flunarizine Hydrochloride Capsules group and high or middle dose of “Hejiezhitong Granule” group were lower than that of low dose of“Hejiezhitong Granule”(P<0.05).Conclusion: 1.when migraine attack,c-fos and c-jun gene expression in brainstem ofmigraine rat model increased significantly.It can be considered to be one of the endogenous mark the launching of neurogenic inflammation.2.“Hejiezhitong Granule” can shorten the time of rats red ear,reduce the number of rats scratching head and climbing the cage.It significantly reduced the number of c-fos and c-jun protein expression in brainstem.So“Hejiezhitong Granule” has some preventive and treatment of effect on migraine.
