Objective: To analyze the honokiol liposome therapeutic effect on vascular dementia and its mechanism.Methods: This experiment by unilateral ligation of the common carotid artery occlusion of vascular dementia animal model making,90 male Wistar rats points to negative blank control group,positive control control group,positive control group,and honokiol liposome injection of high dose group,middle dose group and low dose group.The negative blank control group selected healthy rats can;positive blank control group rats,do not take drug treatment;positive drug control group after making model given irrigation stomach to drug troxerutin aqueous solution 10ml;and honokiol liposome high dose group rats after given intravenous injection of honokiol liposome 0.30ml;middle dose group given intravenous injection of honokiol liposome 0.10 ml;low dose group given intravenous injection of honokiol liposome arterial blood gases and respiratory mechanics were recorded during IPV.For 7 days,during normal eating and drinking water,7 days after the behavioral test,behavior detection after his deathdeath of former 20 min nerve defect score,score immediately after removal of the brain,isolated from the hipocampus in ice bath on the left side,analyze related biochemical detection and fluorescence quantitative PCR,residual into freezing stored in the liquid nitrogen for repeated detection of lung,heart and liver and kidney,and the right of the rats in the formalin solution after pathological and pathological analysis,judgment and treatment effect of magnolol on vascular dementia and its molecular mechanism may exist at the same time,according to the analysis of the high dose group of Honokiol in liver kidney and other organs.The pathological damage that honokiol treatment.Results: Honokiol liposomes of high dose group and middle dose group from the fourth day and sixth day start can significantly shorten the vascular dementia rats in Morris water maze escape latency,positive drug hydroxyetbylrutin control group had significant difference(P < 0.05 or P < 0.01),low dose group is not significant difference.Search space experiments and honokiol liposomes of high dose group and middle dose group can significantly prolong the VD rats in the original safety board around 10 cm swimming distance and the swimming time,and a significant increase in the number of Safety Board(P < 0.05 or P < 0.01).There were significant differences in the control group(or P<0.01),but there was no significant difference between the low dose group and the control group(or P<0.05).After unilateral carotid artery ligation model was made,in hipocampus of rats produced a series of oxidative stress injury,honokiol liposome and the positive drug can treat brain oxidative stress damage,can improve the content of SOD in rat hipocampus,reduce the content of MDA in order to protect the damage by oxidative stress in rat hipocampus.Through the detection of monoamine neurotransmitter in rats,draw and honokiol liposomes and positive drug control group significantly increased in brain tissue of rats with vascular dementia ne,Da,5-HT and 5-HIAA content and the difference had statistical significance(P < 0.05).And honokiol liposomes with high,medium and low dose group,positive drug control group and positive control group can significantly increase e ach content in brain(P < 0.05 or P < 0.01).Fluorescence quantitative PCR results showed(qt-pcr)results show probably by inhibiting Bax expression and enhance bcl-2 expression and decrease the neuronal cel apoptosis,so as to improve the rat learning ability and survival state and play the anti senile dementia.Conclusion: Treatment to ligation of unilateral carotid artery occlusion of vascular dementia rats disease models,honokiol liposome can be significant improvement in vascular dementia rats learning and memory ability and behavior ability.The mechanism may be through increasing the ratio of Bcl-2 / Bax,inhibit the apoptosis of nerve cel aleviate cerebral ischemia on the neurovascular unit damage thereby inhibiting VAD rats in response to oxidative stress,enhanced the antioxidant defense mechanism in brain tissue,improve the ability of learning and memory of the rat model of vascular dementia.
