Huoxue Puzzle Tablets In The Treatment Of Vascular Dementia, Pharmacodynamics And Mechanism Of Experimental Research

Objective:HuoXie YiZhi Tablets has the function of Qi-invigorating and Blood–activating,intelligence-promotion and Orifice-Opening. To investigate the mechanism of its effect on vascular dementia (VD)in animal model.Methods:Using the mouse model with acquired memory disorder resulting from hyoscine and Step-down test,It is suggested that HuoXieYiZhi Tablets can improve the impaired learning and memory induced by scopolamine in mice.The mice and rats model of dementia by ischemia/reperfusion was induced by repeated ligation of bilateral common carotid arteries together with an intraperitoneal injection of sodium nitroprusside. Morris water maze was used to detect the changes of the abilities of learning and memory in rats. To observe the effect of HuoXieYiZhi Tablets on the activity of acetylcholinesterase(AChE)in mouse brain tissue,learning and memory function in rats,the content of oxide (NO),5-hydroxytryptamine (5-HT) in rat brain tissue.And by the And to explore the mechanism of the expression of N-methyl-D-aspartate receptor1(NMDAR1)and brain-derived neutrophic factor(BDNF) in VD model rats.Result:Acute toxicity text showed that maximal tolerance of experimen tal mice to Shizhen xiao Granule was 167.0g/kg/day. It was 104.4 times as the dose in people (adult body weight by 60kg calculated). It is suggested that HuoXieYiZhi Tablets can improve the impaired learning and memory induced by scopolamine in mice. And the protective effects of HuoXieYiZhi Tablets on cholinergic neurons in VD model mice;can improve learning and memory impairment of VD rats,increase the content of 5 HT in biain,can suppress the marked increase of NO caused by insufficient blood to the brain.And HuoXieYiZhi Tablets has the antagonism action of experssion of NMDAR1,promoted the experssion of BDNF.Conclusion:HuoXieYiZhi Tablets has a a very low toxicity.And it can revive the neurologic impairment which was induced by the ischemia/reperfusion model, might have therapeutical effect effective in vascular dementia.
