Purpose: 1.To analyse the characteristic mi RNA expressed differently in human osteosarcoma cells,validate and analyse the mi RNA with markers in clinical samples,screen and capture the mi RNA which is closely related to biological characteristics an d take the mi RNA as the target mi RNA.2.To analyse the regulation and control mec hanism of the target mi RNA makes on the osteosarcoma cells biological characteristic s.3.To analyse the application values of the target mi RNA has in diagnosis and treat ment of osteosarcoma.Method: This research intends to adopt the mi RNA chip to analyse differential expression profile of the mi RNA in G1/S phase,screen and verify the mi RNA with significant difference expression in clinical specimens,select the high consistency in the mi RNA of MG3 and normal osteoblast cells as potential molecular markers for early diagnosis of osteosarcoma,and study the mechanism of the osteosarcoma development features,explore the possibility of using as clinical therapeutic targets.Result: 1.In the cancerous tissue of MG63 cells and the clinical specimens of osteosarcoma,7 mi RNA have consistency differentially expression characteristics,mi R-4500 was significantly higher expression in MG63 cells and osteosarcoma carcinoma tissue.2.The role of mi R-4500 targets may be p53 gene and its signal pathway related genes.3.Cuting down the has-mi R-4500 expression significantly increase the sensitivity of MG63 cells acted on doxorubicin hydrochloride.Conclusion: we through detection of G1/S osteosarcoma cell line MG63 cells of mi RNA expression profiles,screening and in clinical samples to verify the mi R-4500 in osteosarcoma cells in significantly higher expression,inhibition of its expression can increase the sensitivity of MG63 cells to doxorubicin hydrochloride and its target may is p53.We believe that mi R-4500 may have a tumor marker and therapeutic target for anti tumor drug resistance value in clinical application,the specific mechanism remains to be elucidated.
