PART ? Micro RNA-223 and epithelial cell transformation sequence 2(ECT2)expression in the tissue of osteosarcoma and significanceObjective: to study the miRNA-223 and epithelial cell transformation sequence2(ECT2)changes and its clinical significance in the tissues of osteosarcoma.Methods: collect the xinjiang autonomous region people's hospital clinical specimens of 68 cases of bone sarcoma with anhui medical university,use the methodof real time fluorescence quantitative rt-pcr detection osteosarcoma tissues in the specimens of 68 cases of osteosarcoma,edge of tumor tissue,the miRNA-223 expression in normal tissue changes;With real-time fluorescence quantitative rt-pcr and immunohistochemistry methods osteosarcoma tissues in the test specimens of 68 cases of osteosarcoma,edge of tumor tissue and normal tissue in ECT2 expression changes.Results: 68 cases of specimens of osteosarcoma tissue expression of micrornas-223 significantly lower than normal tissue,the tumor edge expression is located in between the two in the organization,and was statistically significant(P < 0.05).All specimens of 68 cases of osteosarcoma tissues ECT2 expression is significantly higher in normal tissue,the tumor edge expression is located in between the two in the organization,and was statistically significant(P < 0.05).Conclusion: the expression of osteosarcoma tissue of the miRNA-223,the expression of ECT2 rise,the miRNA-223 and ECT2 participate in the development of osteosarcoma,there may be a link between both.PART ? Micro RNA-223 in vitro experiments of osteosarcoma cell biology behavior influence researchObjective: to study the miRNA-223 for osteosarcoma cell biology behavior including cell proliferation,cell cycle,apoptosis,adhesion,invasion and the influence of ability to promote angiogenesis.Methods: using liposome method makes the miRNA-223 analog transient transfection into the bone sarcoma cells(MG63),the experimental set up blank control group,negative control transfection group,the miRNA-223 transfection group three groups,fluorescence microscope after transfection transfection efficiency,with real-time fluorescence quantitative rt-pcr detection the expression of micrornas-223 groups of cells and ECT2,proliferation experiment testing groups determined by MTT cell proliferation,flow cytometry instrument testing each cell cycle and cell apoptosis rate,cell adhesion experiment testing each cell adhesion ability,Transwell cell invasion experiment testing each cell invasion ability,small tube forming experiment testing groups of cells to promote angiogenesis ability.Results: the MiRNA-223 transfection group significantly higher,the expression of micrornas-223 ECT2 expression significantly decreased,with statistical significance(P < 0.05);MiRNA223 transfection group of tumor cell proliferation rate significantly decreased,and cell cycle stop before the G1 phase,increased apoptosis rate,cell adhesion and aggressivity obviously decrease,promote angiogenesis ability significantly decreased,with statistical significance(P < 0.05).Conclusion: the MiRNA-223 can effectively inhibit bone sarcoma cell proliferation,adhesion,invasion and the ability to promote angiogenesis,promoting apoptosis and MiRNA-223 is a kind of osteosarcoma suppressor genes,the MiRNA-223 and possible to regulate relations between ECT2.PART ? Micro RNA-223 and epithelial cell transformation sequence 2(ECT2)composite shaft abnormal expression and correlation analysis of the prognosis of osteosarcomaObjective: to study the mirna-223 / ECT2 abnormal expression in osteosarcoma composite shaft to the value of the prognosis of osteosarcoma.Methods: from 2006 to 2009,and xinjiang autonomous region people's hospital of anhui medical university Yi Fu Yuan confirmed 68 cases of patients with osteosarcoma.All patients were given preoperative neoadjuvant chemotherapy,including methotrexate,more gentle than the star,cisplatin,ring ling amide,all give intravenous drugs.Give the surgical removal of the tumor after chemotherapy.And surgery in tumor tissue and tumor tissues around the disease.Get along with all the relevant results to complete the LABS,we with miR-223,ECT2 numerical range based median,miR-223 is 2.50,respectively,ECT2 4.79 will be.MiR-223,ECT2 divided into below and above the median of the median expression,miR-223 low expression group 38 examples,highexpression group 30 cases;ECT2 low expression group of 32 cases,high expression group of 36 cases.On this basis,according to the results of the 68 patients,they can be divided into four groups of 10 cases of miR-223 low/ECT2,28 cases of miR-223ECT2 high/low,22 cases of high/low ECT2 miR-223,8 cases of miR-223 high/ECT2 group.Grouping of postoperative follow-up of patients,all patients before chemotherapy every 3 months 3 years,4 years after chemotherapy once every four to five months,and once every three months to complete CT examination and imaging examination.According to the results of follow-up cure,recurrence and metastasis.Results: follow-up of 5.8 to 36.3 months,an average of 29.6 months,28 cases with metastasis and recurrence,15 cases died during follow-up,the total survival rate was73.53%,3 years 3 years disease-free survival rate was 58.82,four groups of the prognosis of patients with a combination of miR-223 / ECT2 have obvious differenceConclusion: the abnormal expression of miR-223 / ECT2 axis has significant correlation with bone sarcoma with aggressive,have must be related to the prognosis of bone sarcoma,miR-223 low/high ECT2 chemotherapy response is poor,high malignant degree,high recurrence rate.
