ObjectiveColorectal cancer is a serious threat to the health of people. Related studies have shown that Long chain noncoding RNA plays an important role in the development of tumor, in recent years about the long chain non-coding RNA in the colorectal cancer has become the current hot research.Long chain noncoding RNA- PVT1 on chromosome 8 q24. 21, is a highly correlated with the tumor area.Pvt1 knockdown promoted the cell apoptosis, and the expression level of Pvt1 in colorectal cancer cells is higher.Therefore detect colorectal cancer tissue long noncoding RNA Pvt1 expression level, Comparing PVT1 expression level and carcinoma tissue differentiation degree, the relationship between the pathological staging,looking for new colorectal cancer markers and therapeutic targets. MethodsThe first part of the experiment: The colorectal cancer samples from clinical patients, divided into adjacent non-tumor tissue and carcinoma tissues,extract the organization's total RNA, take 5 ul total RNA extraction liquid, using reverse transcription kit to carry on the reverse transcription reaction, synthetic double-stranded complementary cDNA;Using SYBR ? Green Realtime PCR Master Mix kit to quantitatively express the PVT – 1.The second part: The experiment needs 5 u l RT-PCR amplification products for Agarose gel electrophoresis,add 1u l 6 x loading buffer to the mixed sample, 120 v voltage electrophoresis 20 min.Using Baygene BG- gdsAuto520 gel imaging system analyzer ultraviolet detection. ResultPart I: PVT1 expression level in cancerous tissue is 3.1 times than tissue adjacent to carcinoma in all specimen of 76 cases. The purpose gene expression level was statistically similar between the two groups, differentiation of low group is higher than in the middle differentiation group(P<0.05). PVT 1 expression level in cancerous tissue of stage III and stage IV was significantly higher than "zero phase, phase I and phase II" group(P<0.01). Patients with stage II group PVT 1 expression level in cancerous tissue were significantly higher than those with stage I, Patients with stage III group PVT 1 expression level in cancerous tissue were significantly higher than those with stage II, The difference was statistically significant(P<0.01).PVT1 expression level has nothing to do with gender, age.The second part: Gel electrophoresis test confirmed that cDNA content in carcinoma tissue is higher than adjacent tissue. Conclusion1. Pvt1 expression level is higher in the cancer tissue than tissue adjacent to carcinoma.2. Pvt1 expression level is negatively correlated with tissue differentiation types in cancerous tissues.3. Pvt1 expression level is positively related with pathological staging in cancerous tissues.
