Objective:To explore the role and significance of HMGB1 in the expression of IL-33 through the detection of high mobility group box-1(HMGB1)and interleukin 33(IL-33)in patients with allergic rhinitis(AR)and observation their expression in vitro cell culture experiments.Methods:1? We take the turbinate mucosa of 20 AR patients and 10 normal turbinate mucosa cases as control group.We used immunohistochemical methods to detect the expression of HMGB1 and IL-33 in the nasal mucosa.And we make correlation analysis between these two cytokines.2? We culture the human nasal mucosa epithelial cell in vitro and use the house dust mite allergen(Derp1)to stimulate the cell.And then we detect the expression of HMGB1 and IL-33 in cell by using fluorescent quantitative PCR method.3? Todetect the expression of HMGB1 and IL-33 through enzyme-linked immunosorbentassay(ELISA).Results:1? The immunohistochemistry showed that the expression of HMGB1 and IL-33 in nasal mucosa of patients with AR was higher than that in normal group,and the difference is statistically significant(P ? 0.05);HMGB1 protein was mainly expressed in the nuclei of epithelial cells in nasal mucosa,and IL-33 was both expressed in nucleus and cytoplasm.And the expression of IL-33 was positively correlated with the expression of HMGB1.2? In vitro cultured epithelial cells of nasal mucosa,fluorescence quantitative PCR detection demonstrated that the expression of HMGB1 and IL-33 m RNA in AR group was higher than that in normal group,and the difference was statistically significant(P < 0.05);The expression of HMGB1 and IL-33 m RNA in stimulated group was higher than that of non-stimulated group and the difference was statistically significant(P < 0.05);The expression of HMGB1 and IL-33 m RNA increased gradually with the increasing concentration of Derp1;And the expression of HMGB1 and IL-33 m RNA increased significantly after 2h of stimulation and was highest in 24 h.3? The results of ELISA showed that the expression of HMGB1 and IL-33 in stimulated cells was higher than that in non-stimulated group and the difference was statistically significant(P < 0.05);The expression of HMGB1 and IL-33 increased gradually with the increasing concentration of Derp1;The expression of HMGB1 and IL-33 increased significantlyafter 2h of stimulation and was highest in 24 h.Conclusions:1? The expression of HMGB1 and IL-33 were both increased in AR.2? HMGB1 may promote the expression of IL-33 in AR.
