Background Esophageal carcinoma( EC)is the eighth leading cause of cancer-lelated mortality worldwide and the most common carcinoma in our country. EC has two main subtypes, esophageal squamous cell carcinoma(ESCC) and esophageal adenoma-carcinoma(EAC). ESCC is the predominant histological type,which has a wide distribution,especially in Henan, Hebei, Shanxi and other provinces. Although lots of studies focus on the early diagnosis,early treatment have been conducted to explore the mechanism of ESCC, the etiology and pathogenesis of ESCC remain unclear, without breakthrough for the early diagnosis and early treatment. It is still a threat to life expectancy and the middle-late patients' overall 5-year survival ranging from 10%- 15%. The key for early diagnosis,early treatment and the higher survival rate is the study for the mechanism of tumor developing. Long non-coding RNA is a complicated RNA which has no or few function in the prcocess of coding RNA.It is transcribed in most eukaryotic genomes, participated in the process of X chromosome silence, genomic imprinting, chromatin modification, transcription activation, transcription interference, nuclear transport and other important regulation progresses. Previous studies showed that Lnc RNA H19 is a mature production of gene H19 and has a high expression level in the tumor tissues such an bladder cancer, kidney cancer,gastric cancer, pancreatic cancer and so on. It plays a major role in the process of proliferation,apoptosis,invasion,and metastasis in tumor cell. In this study, q RT-PCR is applied to test the expression level of ESCC and the adjacent tissues from the patients of ESCC patients who received the radical surgery. Furthermore, we analyzed the relationship between the expression level of Lnc RNA H19 and the clinical pathological characteristics and prognosis of ESCC patients.Aim To explore the expression status and prognostic significance of long noncoding RNA(Lnc RNA) H19 in ESCC patients who received radical resection.Methods Quantitative real-time polymerase chain reaction(q RT-PCR) was applied to detect the expression level of Lnc RNA H19 in carcinoma tissue and paired adjacent tissue samples from 68 ESCC patients who received radical surgical resection. The correlation of the relative expression level of Lnc RNA H19 with clinical-pathological features and prognosis were also analyzed.Results 1. Lnc RNA H19 was upregulated in70.6%(48/68)ESCC tissues and the relative expression of Lnc RNA H19 was significantly higher in ESCC tissues compared with adjacent tissue samples(P=0.023). 2.The relative expression level of H19 Lnc RNA was not associated with sex, age, T stage, lymph node status, histological grade, TNM stage and so on(P>0.05). 3.Kaplan-Meier survival analysis showed that patients with high expression of H19 had a shorter DFS and OS than those with low Lnc RNA H19 expression(P=0.026,P=0.040).Multivariate analysis showed that the HR of Lnc RNA H19 expression was 0.515( 95%CI:0.287-0.924, P=0.026) for DFS and 0.504 (95%CI:0.265-0.961, P=0.038)for OS, respectively.Conclusion 1. Lnc RNA H19 is upregulated in the ESCC. 2.Our results indicate that high expression level of Lnc RNA H19 may associate with poor prognosis of ESCC, and it may be a prognostic indicator for ESCC patients who received radical resection.
