Objective: To study the effects and significance of hypoxia on the expression of matrix metalloproteinase 9(MMP9) and hypoxia induced factor 1(HIF-1?) of 786-O renal carcinoma cell line in m RNA levels and protein levels.Methods:786-O renal carcinoma cell line was respectively incubated with 150?mol/L Co Cl2 for 8hours,16 hours and 24hours;Western blot and real time polymerase chain reaction(RT-PCR) were used to detect protein and m RNA levels of HIF-1? and MMP9.Results:HIF-1? and MMP9 have a certain degree expression in protein and m RNA levels in normoxia state; The m RNA and protein expression levels of MMP9 was up-regulated in hypoxia group(P <0.05), and the levels was increased with prolonged time; The changes of m RNA level of HIF-1? didn't have significant difference(P >0.05),but the protein level of HIF-1? was increased significantly(P <0.05).Conclusion: The expression of HIF-l? m RNA was not significantly increased, however the expression of protein increased significantly in hypoxia.It means that the expression of HIF-l? was regulated at translation level in hypoxia. The expression of MMP9 significantly increased at both transcription level and translation level in hypoxia. HIF-l? and MMP9 may have important effects in the occurrence, development, invasion and metastasis of renal cell carcinoma.
