Abstract/Summary:
Objective 1. To investigate the murine double mimut 2(MDM2) and ribosomal protein L23(RPL23) expression in gastric cancer and paracancer,explore their biological significance in the development of gastric cancer. 2. analyze the relationship between the expression of RPL23、MDM2 andclinicopathologic factorsof gastric cancerpatients. 3. Discussion the correlation of MDM2 and RPL23 expression in gastrictissue 4. To explore RPL23 and MDM2 expression in different TCM Syndromes.Simultaneously,we discussethe relationship between TCMSyndrome anddepth of tumor invasion、lymph nodemetastasis. To provide a theoretical basis for the relationshipbetween TCM Syndrome and RPL23 and MDM2.Methods Selection 100 cases from October 2014 – October 2015 in Wuhan General Hospital, Guangzhou Command of PLA surgery patients, pathologically diagnosed with stomach cancer. We selected the medical records of patients,respectively with reference to “the traditional Chinese medicine oncology”(Editor: Zhou Dai Han, the Chinese Medicinepublishing, 2011). The gastric cancer is divided into disharmony of liver and stomach,Qi stagnation type,phlegm coagulation, deficiency of spleen and kidney. Clinicopathological factors including age, sex, smoking, depth of tumor invasion(T stage), tumor size, lymph node metastasis. The 30 cases in control group were obtained from adjacent non-cancerous tissues. The expressions of MDM2 and RPL23 were detected by immunohistochemistry in two groups. The result was analyzed by statistical SPSS19.0.Results 1、The expressions of RPL23 and MDM2 are different gastric cancertissuesandadjacent normaltissues.The positive expression rates of RPL23 in gastrictissues was significantly lower than that in control group(24% vs 63.3%, P<0.05), while MDM2 expression was significantly increased(61% vs 40%, P<0.05). 2、The result showed that the stainingof RPL23 andage(χ~2 = 2.268, P = 0.132),gender(χ~2 = 2.722, P = 0.099),tumor size(χ~2 = 0.305, P = 0.581),depth of tumor invasion(χ~2 = 1.046, P = 0.306)unrelated. But it has asignificantcorrelation between RPL23 andlymph nodemetastasis(χ~2 = 13.193, P = 0.00),smoking(χ~2 = 4.346, P = 0.037). 3、MDM2expression andage(χ~2 = 0.408, P = 0.523),gender(χ~2 = 0.087, P = 0.768),tumor size(χ~2 = 0.17, P = 0.68),depth of tumor invasion(χ~2 = 0.248, P=0.618), smoking(χ~2 = 0.583, P = 0.445)irrelevant. MDM2 and lymph nodes(χ~2 = 4.691, P = 0.03)haveobvious correlation. 4、The expression of MDM2 and RPL23 in gastric cancers was negativecorrelated(r =-0.245, P = 0.014) 5、gastric cancer patients with RPL23,MDM2 protein expression and expression levels between different TCM syndromes of gastric cancer, there is a significant difference(P<0.05) 6、The expression of RPL23 protein levels in different typing from strong to weak: disharmony of liver and stomach>Qi stagnation type>phlegm coagulation>deficiency of spleen and kidney(37.21%>21.05%>11.11%>9.09%). 7、The expression of MDM2 protein levels in different typing from weak to strong: disharmony of liver and stomach<Qi stagnation type<phlegm coagulation<deficiency of spleen and kidney(37.21%<63.16%<85.19%<90.91%). 8、Depth of tumor invasionand TCM syndromeof gastric cancerhas statistically significant correction(P<0.05),while thelymph nodemetastasis and TCM irrelevant(P> 0.05)Conclusion 1、MDM2 and RPL23 has a certain relevance with gastric cancer. 2、RPL23 expression in early gastric cancer is higher,but MDM2 in advanced gastric cancer is higher.It can indirectly speculate gastric degree by the expression levels of both genes. 3、MDM2 upregulation, RPL23 downregulation, gastric Empirical more obvious, the illness is more complex,the worse the prognosis.The two proteins may be thematerial basis for the formation ofdifferent TCM syndromes.
