Objective:With the growing problem of aging population, some diseases associated with aging, such as Alzheimer's disease(AD) have been paid more attention gradually. It's extremely important to strengthen the research on etiology and pathogenesis of Alzheimer and find effective drug of prevention and treatment in clinical value and practical significance. The production of phlegm runs through the whole process of the development of Alzheimer's and is the main factor in the disease worsening. Therefore, this study discussed on the treatment of Alzheimer's from phlegm, using the method of behavioral, electrophysiological and biochemical to explore the research of Hutan Prescription on synaptic plasticity and oxidative stress in AD mice. It provides theoretical and experimental foundation for further study of the treatment of Alzheimer in Chinese medicine.Method:1. Theoretical discussion:Collate the literature between sputum and Alzheimer, explore the production of phlegm and the theoretical basis on the treatment of Alzheimer's from phlegm.2. Experimental study:Using APP/PS1 transgenic mice as animal model of AD, study the effect and mechanism of Hutan Prescription which is Ditan Tang and Yizhi Wendan Fang in Alzheimer. Detect the learning and memory ability of mice by the behavioral indicators such as morris water maze and step-down test. Induce the LTP of mice in hippocampal with nerve electrophysiological testing. Detect the content and vitality of SOD, MDA, T-AOC in mice brain by colorimetric method.Result:1. Morris water maze test showed that compared with the control group, the platform latency and total swimming distance in model group were significantly longer(P <0.01), the number of crossing platform and target quadrant time were significantly reduced(P<0.01); compared with the model group, the total distance swimming,the platform latency and the first time to platform of Yizhi Wendan group and Ditan Tang group were reduced(P <0.01, P <0.05), the number of crossing platform and target quadrant time were increased(P <0.01, P <0.05).2. The step-down test showed that compared with the control group,the number of error times in model group increased and incubation period shortened significantly(P <0.01); compared with the model group, the number of error times in western medicine group and Yizhi Wendan group reduced(P <0.01, P <0.05), the latency of western medicine group was prolonged(P <0.01).3. The results of hippocampal LTP induction showed that control group can induced a stable LTP after giving a high-frequency stimulation, however, the model group could not be successful using the same method(P <0.01); compared with the model group, the slope of EPSP in Yizhi Wendan group was increased(P <0.01), but the Ditan Tang group did not improved significantly(P> 0.05).4. The results of SOD, MDA, T-AOC show showed that compared with the control group, the content of SOD in the model group was reduced and MDA increased significantly(P <0.01); compared with the model group, the content of SOD and T-AOC in other groups increased and MDA reduced(P <0.01, P <0.05).Conclusion:1. Alzheimer's disease is located in the brain and is closely related to dysfunctional internal organs like heart, liver, spleen and kidney in Chinese medicine. The inherent mechanism of dementia is the virtual failure of Five organs especially kidney deficiency.Phlegm is an important factor in accelerating the occurrence of Alzheimer. Both affect and reinforce each other which formed the vacuity of actual situation mixed. The production of sputum runs through the whole process of the development of Alzheimer's and is the main factor in the disease worsening. Many kinds of internal and external factors can affect the production of phlegm, such as eating habits, daily life, emotions, climate and so on. It's the result of comprehensive effect of internal organs, qi, blood and body fluids. The most closely related to the phlegm generated is spleen and kidney, followed by the lung and liver.2. Yizhi Wendan Fang and Ditan Tang can improve learning and memory ability of the APP/PS1 transgenic mice. It is possible to effectively improve the learning and memory impairment of APP/PS1 transgenic mice through improving the function of synaptic plasticity.3. The oxidative stress involved in the pathogenesis of AD.Yizhi Wendan Fang and Ditan Tang can increase the content of SOD and T-AOC and decrease MDA in APP/PS1 transgenic mice. It suggests that its neuroprotective effect may be related to the antioxidant effects.
