Objective: To investigate whether mir-132-3p targets SOX4 to inhibit the proliferation and migration of Hep G2 cells.Methods: Hep G2 cells were divided into control group without any treatment, mimics transfection group and hsa-mir-132 mimics transfection group.The expression of mir-132-3 p and SOX4 m RNA in hep G2 cells were detected by real-time fluorescent quantitative PCR, SOX4 expression was detected by western blot; Hep G2 cell proliferation was determined by MTT at 24 h, 48 h, 72 h respectively; Flow cytometry detected cell apoptosis and the changes of cell cycle; Migration was observed by Scratch test and cell invasion was detected by Transwell.Results: 1. Real-time fluorescent quantitative PCR results showed that mir-132-3P expression in hsa-mir-132 mimics transfection group was significantly higher than that in control group and mimics transfection group(3.001±0.218 vs 0.930±0.102 and 0.910±0.088,P < 0.05). SOX4 m RNA expression in hsa- mir-132 mimics transfection group was outstandingly lower than that in control group(0.388±0.114 vs 0.894±0.156 and 0.918±0.157, P < 0.05). 2. Western blot results displayed SOX4/GAPDH value in hsa-mir-132 mimics transfection group was obviously lower than that in control group and mimics transfection group(0.180±0.045 vs 0.647±0.032 and 0.663±0.032, P < 0.05). 3. MTT results showed that cell proliferation rates in hsa-mir-132 mimics transfection group at 24 h, 48 h, 72 h after transfection were apparently lower than that in mimics transfection group(91.77%,84.55%,76.68% vs 99.89%,99.94%,100.56%, P < 0.05) and control group(100%). 4. Flow cytometry results indicated apoptosis rate in mir-132-mimics transfection group was remarkably higher than that in control groupnd and mimics transfection group(18.79%±0.44% vs 6.83%±0.21% and 6.01%±0.25%,P<0.05). 5. Scratch test results demonstrated the number of migrated cells in mir-132 mimics transfection group was evidently lower than that in control group and mimics transfection group after transfection for 24 h( 41±19 vs 232±38 and 225±28, P<0.05); Transwell results declared the number of trans-membrane cells in mir-132 mimics transfection group was prominently lower than that in normal control group and mimics transfection group(29±2 vs 52±2 and 51±1,P<0.05).Conclusion : mir-132-3p overexpression could inhibit the proliferation and migration of Hep G2 cells via down-regulating SOX4 gene expression.
