| Objective:To investigate the effects of Icariin(ICA) on the proliferation of human ectopic endometrium cells and the expression of MMP-2、MMP-9 protein and mRNA,in order to provide experimental and theoretical basis for the treatment of endometriosis. Method:Different concentrations(0, 25, 50, 100, 200μg/ml respectively) of ICA were added to the medium and human ectopic endometrium cells were further cultured for 24 hours,MTT assay was used to detect the proliferation effects of different concentrations of ICA on human ectopic endometrium cells. RT-PCR and ELISA assay were used to measure the different levels of MMP-2、MMP-9 protein and mRNA expression in human ectopic endometrium cells which were processed by ICA(200μg/ml) after 24 h. Result:1, the in vitro experimental results showed that EMs cells abnormally increase the expression of MMP-2, MMP-9 protein and mRNA than the normal endometrial cells, the difference was statistically significant, P <0.05.2, MTT assay showed ICA( 25, 50, 100, 200μg/ml) can inhibit the proliferation of EMs cells on different levels.The inhibition rate of ICA with the concentration of 25μg/ml has no statistically differences,while the other groups remaining statistically significant differences,P <0.05, which may indicates the ICA have a dose-dependent manner on the inhibition of EMs cells.3, After cultivating the experimental EMs cell group with the ICA(100μg/ml) 24 h, the expression of the MMP-2, MMP-9 protein and mRNA levels decreased significantly than the group without the ICA.The difference has statistically significance, P <0.05. Conclusion:ICA can inhibit the proliferation of EMs cell and drive the expression of MMP-2, MMP-9 protein and mRNA of EMs cell down, which may weaken the adhesion and invasion of the EMs cell. ICA can be a potential new drug for the treatment of EMs. |
