Systemic lupus erythematosus(SLE)is a kind of autoimmune disease involving multiple systems,which is mainly characterized by the activation of the complement system and the deposition of immune complexes.Due to the pathogenesis of this disease is not clear,in recent years,with the continuous development of the sugar group,confirmed that the immune globulin glycosylation modification involved in the pathogenesis of autoimmune diseases and induced immune tolerance.Objective: To investigate the correlation between the changes of serum N Ig G-Fc sugar chain structure and the incidence of SLE in patients with systemic lupus erythematosus.Methods:(1)fasting venous blood was collected from 50 SLE patients and 26 healthy pregnant women,and the supernatant was stored in-80??Collect all the general condition of patients with systemic lupus erythematosus(SLE),clinical features ?laboratory results and according to the SLE disease activity index SLEDAI-2000 of 50 patients with SLE clinical staging score less than or equal to 4 points as the stationary phase,score is greater than or equal to 5 is divided into active stage.(2)the serum total immunoglobulin was separated and purified with proteing G,after SDS-PAGE validated Ig G,the purified Ig G with PNGase digestion.After further enriched and purified by N-linked sugar chain and by lectin microarray can be specific to affinity immunoglobulin(Ig G)surface sugar chain characteristics,laser scanning measuring lectin signal determination of SLE(stationary phase,active phase)and pregnant women with IVIG Ig G glycans on the surface structure(3)the use of statistical methods to compare theSLE(active and inactive)?normal pregnancy and IVIG glycosylation abnormalities and analyze the correlation of SLE active stage and stable stage patients serum sugar chain structure change with lupus disease activity and renal damage experiment index.(4)make use of characteristic of lectin agglutination to specific affinity of sugar chain immunoglobulin(Ig G)surface properties,laser scanning detection signals was developed for the determination of SLE,pregnant women with IVIG Ig G sugar chain structure of the surface.Results:(1)the content of G2 galactose,the core fucose,salic acid in SLE were significantly lower than the IVIG and normal pregnancy,the difference was statistically significant(P<0.05),G0 galactose and high mannose content was significantly higher than that in IVIG and normal pregnant women,the difference was statistically significant(P<0.05),and there was no significant difference at G1 galactose(P>0.05)(2)In active SLE patients the serum Ig G surface of type G2 galactose and sialic acid content compared with inactive SLE patients decreased significantly,the difference is statistically significant(P < 0.05),while G0 galactose and mannose content compared with active SLE was significantly higher,the difference is statistically significant(P < 0.05),the active stage of SLE core sugar level in Dunaliella salina and stationary phase difference no statistical significance(P > 0.05).The Ig G of patients with SLE the content of G0 galactose(r=0.704 P<0.000)and highmannose(r=0.731 P<0.000)was positively correlated with SLEDAI score,G2 galactose(r=-0.792 P<0.000)was negatively correlated with SLEDAI score and there was no significant correlation between salivary acid and SLEDAI score(P>0.05 r=0.004).(3)Analysis of the relationship between the experimental parameters of renal injury in patients with SLE during the period of disease activity and restperiod.Compare with the stable phase There was significant difference in the level of ds-DNA antibody in patients with SLE active phase(P<0.000),24 h urinary albumin in the SLE activ phase and the stationary phase difference has statistical significance(P<0.000),There was significant difference in ?2MG between activity and stationary phase(P<0.000).Conclusion:(1)Compared with normal pregnant women and IVIG,The glycosylation of Ig G in SLE patient is abnormality.It is suggested that the changes of the structure of sugar chain on the surface of Ig G are related to the pathogenesis of SLE.(2)There was a significant difference in the glycosylation of Ig G with SLE in patients with stable phase of SLE,suggesting that the glycosylation of Ig G was related to the activity of SLE.The levels of serum Ig G in SLE patients were correlated with SLEDAI,C3,C4 and CPR,which suggested that Ig G was correlated with SLE disease activity.(3)In SLE active stage,the levels of ds-DNA antibody,24 h urinary protein and ?2MG were significantly higher than those in the rest period.Ig G surface sugar chain and ds-DNA antibody,24 h urinary protein,beta 2-microspheres have a correlation,suggesting that the changes in the structure of the Ig G surface of the sugar chain and damage to the kidney function are correlated.
