| BRH is a common disease of the elderly men, which has become one of the major diseases to affect the health and life’s quality of older persons. It is mainly because of the sex hormones in the body metabolic disorders, which results in varying degrees of prostate volume increase and fibrous tissue hyperplasia and destroy the normal glandular structure, then cause a series of the dysfunction. In treatment, drugs and surgery combined therapy are often applied. Surgery is the most direct and effective means of treatment, generally using laser technology, heat treatment, support and balloon expansion. In drug treatment,5α-reductase inhibitor and α1-blockers are used more. In spite of a significant effect of these drugs, but there are certain side effects. Therefore, we should study the more effective and more safer treatments. TCM clinical treatment of benign prostatic hyperplasia, which often contain Vaccaria and Salvia, and these prescriptions are promoting blood circulation to remove blood stasis, diuretic effects and so on. Clinically for the treatment of elderly prostate hyperplasia, urination, dripping net, women’s blood stasis amenorrhea, high permeability, female infertility and so on. In this paper, three kinds of clinical experience in traditional Chinese medicine compound containing the vaccariae(WDD, WDDBW and WDDBWYM) go on animal drug screening. On the efficacy significant WDD will study in vitro and in vivo mechanism of action. The following main results achieved:(1) Efficacy StudyBy subcutaneous injection of testosterone propionate hormone-induced BPH hyperplasia model, which goes on drug screening with different Chinese herbal compound, WDD’s different extraction methods and the active sites of WDD’s aqueous extract. To observe the drug inhibitory effect and evaluate the efficacy of drug on anti-BPH. Experimental results show that the water extract of WDD have the significant efficacy, mainly active ingredients concentrated in macroporous resin 30% -60% ethanol parts and inhibit prostatic hyperplasia effect is obvious.(2) Studies on the mechanism of WDD extractionDetermination the contents of PSA, ACP, T and DHT in the serum after rat gastric drug delivery and using ELISA; Determination the contents of its DNA by using phenol method and UV absorption method; Determination the contents of WDD water extraction’s two main active extracts (Sal-B and Vaccaria) by using high performance liquid chromatography(HPLC) and detect the changes of WPMY-1 GNHu36 cells proliferation, cycle and apoptosis by using MTT and flow cytometry technique. The system research that the mechanism of WDD’s mainiy active ingredient (Sal-B and Vaccaria) inhibite prostatic hyperplasia. Experimental results show that the WDD’s water extraction can inhibit DNA synthesis, reducing the contents of PSA, ACP (P<0.01) and T (P<0.05) in the serum; WDD’s water extraction contains Sal-B and Vaccaria and the content of Sal-B is 1.25%, the content of Vaccaria is 0.09%; Sal-B at lower concentrations can significantly inhibit WPMY-1 cells proliferation, increase the number of cells in G2 phase and S phase, which significantly induces early apoptosis/late apoptosis and total apoptosis at a concentration of 100μg/ml and 300 μg/ml. Suggesting that Sal-B may be main efficacy component to suppress prostatic hyperplasia.In summary, the WDD’s water extraction has a significantly inhibit prostatic hyperplasia and the active ingredients concentrated in the 30%-60%ethanol parts, by inhibiting the DNA synthesis of animal prostate and reducing the contents of the PSA、ACP、T to achieve the role of inhibition and treatment of benign prostatic hyperplasia. Salvianoid acid-B may mainly be active ingredient by inducing cell apoptosis and cell cycle arrest to inhibit cell proliferation, eventually play against prostatic hyperplasia. |
