Objective: With the development of modern industry,air pollution and the social the aging of the population increasing,the incidence of lung cancer continues to increase worldwide,already the first systemic malignant tumor incidence.The occurrence of lung cancer is a very complex process,including Shared by a variety of oncogenes and tumor suppressor genes involved in regulation.Lung cancer pathogenesis is very complex,among them,the signal transduction of survival for lung cancer occurrence,development process plays a vital role.Currently,oncogene and tumor suppressor genes influence on the occurrence of tumor and prognosis is becoming a hot spot of research.This study selected cancer gene AKT1 and suppressor PDCD4 as the research target,adopts the immunohistochemical Streptomyces avidin peroxidase connection(SP)method to detect both in lung cancer tissue and normal bronchial and alveolar tissue expression,to explore the role of both in non-small cell lung cancer(NSCLC).Methods: 65 cases of non-small cell lung cancer after surgery,30 cases of alveolar tissue and 30 cases of bronchial stump tissue were enrolled in the study,used immunohistochemistry method to detect AKT1 and PDCD4 in NSCLC group and Dual control group and analyzed its relationship with the clinical and pathological data.1 Lung cancer group and normal control group from June 2011 to June 2014,65 cases of thoracic surgical resection in our patients with non-small cell lung cancer(has not been preoperative radiotherapy,chemotherapy),39 cases of men and women in 26 cases,aged 42 ~ 76,mean age 59.77 + /-9.46 years old.Match each patient's age and gender(all P > 0.05).Grouping: non-small cell lung cancer group,65 cases(surgical removal of the tumor tissue);Every 30 cases of normal control group(ipsilateral distal alveolar and bronchial lung cancer normal tissue);In non-small cell lung cancer group,lung squamous carcinoma 37 cases,28 cases of patients with lung adenocarcinoma;Differentiation carcinoma in the poorly differentiated carcinoma in 11 cases,32 cases of well-differentiated carcinoma 22 cases;TNM staging 25 cases of phase ? and phase ? 12 cases,20 patients with stage ? and ? 8 cases.2 Routine histological processing: all specimens were fixed in 10% formalin solution,embedded in paraffin and sliced,HE staining was for pathological examination.Tumor histologic classification reference 2004 edition of the world health organization(WHO)lung tumor classification criteria.3 Immunohistochemical detection:Immunohistochemistry: tissues were paraffin-embedded and sliced continuously with 3.5um ~ 4um thickness,oven 60? ~ 65 ? overnight,dewaxed,citric repair liquid boiling to repair,used SP immunohistochemical staining according to immunohistochemistry kit manual steps strictly,colored with DAB,hematoxylin staining,With known AKT1 and PDCD4 protein positive biopsy as positive control,resistance to PBS liquid instead of one as a negative control.4 Interpretation of the results: used double-blind method to observe results under a microscope.The nucleus and cytoplasm tan is positive,negative not shaded.Positive determination was judged by positive cells and staining intensity.5 Statistical treatment: The experiment data were used by SPSS 16.0 software and analyzed by c2 test,P <0.05 was considered the difference was statistically significant.Results:1 The expression of AKT1 in NSCLC Group was significantly higher than Dual control group(P<0.05).2 Expression of AKT1 protein in NSCLC was significantly related with tumor differentiationlymph,node metastasis and TNM staging(P<0.05),but was no statistical significance in groups of gender,age,tumor size and histological type(P>0.05).3 The expression of PDCD4 in NSCLC Group was lower than Dual control group(P>0.05);4 Expression of PDCD4 protein in NSCLC was significantly related with tumor histological type(P<0.05),but was no statistical significance in groups of gender,age,tumor size differentiation,lymph node metastasis,and TNM staging(P>0.05).5 Expression of AKT1 and PDCD4 in lung adenocarcinoma tissue was negatively correlated(r =-0.436).Conclusion: The onset of AKT1 and PDCD4 in non-small cell lung cancer play a role in the process,AKT1 as a oncogene expression in non-small cell lung cancer,prompt AKT1 in non-small cell lung cancer occurrence process effect on promoting cancer,And AKT1 protein expression and lung cancer associated degree of differentiation,lymph node metastasis and TNM stages,The lower level of the higher expression prompt differentiation of lung cancer,malignant degree is higher,the worse prognosis,For clinical individual therapy has a certain guiding significance.
