Objective:PI3K blockers as positive control group,we investigate the mechanism of preventing ulcerative colitis associated carcinogenesis(UCAC)by Jian-pi-qing-re-huo-xue decoction(JPQRHX decoction)depending on observing the effect of treatment with Jian-pi-qing-re-huo-xue decoction on SW480 cell proliferation,cycle,apoptosis and the expression of(3-catenin.Method:Using the method of serum pharmacology,JPQRHX decoction drug serum was prepared,and the components of serum was detected by high performance liquid chromatography(HPLC).In vitro,human colon cancer cell line SW480 were divided into a blank group,an intervention group and a control group,which were incubated with fetal calf serum,different concentrations of serum containing Jian-pi-qing-re-huo-xue decoction,or serum containing LY294002 for 24 hours,respectively.The proliferation of SW480 was detected by MTT.The cell cycle and apoptosis of S W480 cells were detected by flow cytometry,and the protein expression of?-catenin was assayed by laser scanning confocal microscope and real-time PCR.Result:The effective components of Jian-pi-qing-re-huo-xue decoction are notoginseng R1?ginseng saponin Rgl and glycyrrhizic acid.The inhibitory rate in intervention group is 2.55%,4.90%and 16.30%respectively.The inhibitory rate in control group is zero.The comparison between intervention group and control group is obvious(P<0.05).The inhibitory rate in LY294002 group is 63.70%.Compared to the blank group,intervention with JPQRHX decoction significantly increased the percentage of cells at S phase and decreased at G1 phase in a concentration-dependent manner(P<0.05).But control group increased the percentage of cells at G1 phase and decreased at S phase compared to the intervention group(P<0.05).Treatment with JPQRHX decoction and control group(14.65±2.54%)increase cell apoptosis compared to the blank group(2.52±0.06%,P<0.05).The different dose of medicated serum increased rate of cell apoptosis was 4.07±0.13%,5.45±0.15%,6.72±0.08%,respectively.Compared to the blank group,intervention group significantly down-regulated the expression of(3-catenin in the cell nucleus(P<0.05).Conclusion:JPQRHX decoction can inhibit SW480 cell proliferation and block SW480 cell cycle at G1 phase,induce cell apoptosis,and have the ability of prevent and treat the colon cancer.Its mechanism possibly by regulating PI3K signal pathway,and altering the expression of?-catenin in the cell nucleus.
