Objective To observe the influence of Polygonatum Sibiricum Polysaccharide( PSP) for angiogenesis responses mediated by Notch1 on acute myocardial infarction in rats and explore the protective effects and mechanisms, and to provide experimental datas for the application of clinical drug development and healthy care products.Methods The AMI models were established by closing the left anterior descending coronary artery with SD rats, sham group wearing without ligation, then the successful model rats were randomly divided into model group, positive group(Shexiang protect myocardial pills, 12.2 mg·kg-1·d-1), PSP low-dose group(225 mg·kg-1·d-1),middle-dose group(450mg·kg-1·d-1), high-dose group(900 mg·kg-1·d-1),10 rats in each group. Gastric infusion one time every day for 28 days. ECG S-T were measured before surgery and immediately after surgery, 1 3, 7, 10, 14, 21, 28 d in each group.After the last administration, 3.5% chloral hydrate anesthetized rats, exsanguinated from abdominal aorta and serum was separated, the concentration serum CKMB, LDH, IL-6, TNF-?,NF-?B and GSH-PX,MDA levels were measured by ELISA; myocardial tissues were fixed with 4% paraformaldehyde, embedded in paraffin, sliced, and morphology was measured by HE staining; myocardial tissues fixed with 4% paraformaldehyde are dehydrated with 30% sucrose solution, frozen sections, measured myocardial tissue CD34 by immunofluorescence staining, counted the number of microvascular( MVC)and Notch1?Hey2 by Western-blotting.Results1.The surface of left ventricular was whitening, severe ischemia, and significanthypertrophy in model group, the drug therapy groups were ruddy. Compared with sham group, model rats were successful because the ST segment was significantly rised after operation.Compared with model group, PSP inhibited ST segment of myocardial infarction(P<0.01 or P<0.05).2. Compared with model group, myocardial cell swelling and muscle bundle disordered were lesser, inflammatory cell infiltration, necrotic cells were decreased from pathology testing by HE.3. Compared with model group,serum levels of CKMB ?LDH decreased in different degrees(P<0.05 or P<0.01);the serum levels of NF-?B?IL-6?TNF-??MDA dropped significantly in the drug therapy groups(P < 0.05 or P < 0.01), the serum level of GSH-PX was significantly higher in high and medium groups(P<0.05 or P<0.01).4.Compared with model group, MVC of treatment groups were increased significantly(P<0.05 or P<0.01);CD34 antibody positive were little in the sham and model group, while the expression of each administration group were significantly higher in infarcted area, endothelial cells arranged in cords, and neovascularization; the expression of Notch1,Dll4,Hey2 were significantly increased in different degrees in drug groups(P<0.05 or P<0.01).Conclusion1.PSP can improve the degree of myocardial injury on AMI model in rat,reduce ST segment for acute ischemia-induced.2.PSP can effectively reduce serum levels of CKMB, LDH, NF-?B, IL- 6, TNF-?, MDA,increase GSH-PX content, reduce the expression of Notch1 Dll4 and Hey2 proteins in myocardial tissue,reduce inflammation and oxidative stress, promote angiogenesis in the ischemic area and repair ischemic myocardium.
