ObjectiveThe study is based on feed with high fat and high sugar, then the rats were inject with STZ by enterocoslia to set up type 2 diabetec model. We want to explore the possible protective effect and mechanism of Polygonatum sibiricum Polysaccharide on myocardial inflammation in diabetic rats.MethodsThere were 50 healthy male SD rats. Chose 40 rats were feeded on high fat and high sugar for about one month and after a month given tail vein injection with Streptozotion of 35mg/kg to establish type 2 diabetes model. However the others were injected with equal volume of citric acid buffer as normal control group. After fasted for 72 hours, cutted the tails to text blood glucose. When the blood glucose level is greater than or equal to 16.7mmol/L, it can be recognized as diabetes model. Then randomly divided into diabetes model group, PSP low (250mg/kg), medium (500mg/kg) and high (1000mg/kg) dose group. According to the PSP dose of 250mg/kg?500mg/kg?1000mg/kg irrigation stomach for about 12 weeks. During the irrigation keeping observe the rats situation. Use the sannuo blood glucose meter text blood glucose level; Biochemistry examine the serum lipid content; BL-420F biomedical analysis system detection the rats heart function; Hematoxylin and eosin staining observe the myocardial tissue pathology change; Immunohistochemistry method and western blot protein mark method to determine the heart tissue of Toll-like receptor-4 (TLR4), Nuclear Factor-KB (NF-?B) and Macrophage Migration Inhibitory Factor (MIF) protein expressions.ResultsCompared with normal group, the model group rats were in poor condition. It seems to be more drink, polyphagia. The rats reactions were not flexible; The eye bolls were mostly change into white; contrast with diabetes group, the phenomenons in the dose of PSP 250mg/kg,500mg/kg and 1000mg/kg groups were improved to some extent;Compared with normal group, the blood glucose in diabetes group were decreased significantly (P<0.01); contrast with diabetes group, the blood glucose levels in PSP groups of 250mg/kg,500mg/kg and 1000mg/kg dose were decreased (P<0.05 or P<0.01);In the diabetes group TG, TC, LDL increased significantly (P<0.01), and HDL reduced significantly (P<0.01); while in the group of PSP 250mg/kg, 500mg/kg and 1000mg/kg TG, TC, LDL were reduced and HDL developed (P<0.05 or P<0.01);Compared with normal group, the ±dp/dtmax in diabetic group reduced significantly (P<0.01), contrast with diabetes group, the ±dp/dtmax in the dose of PSP groups were increased (P<0.05 or P<0.01);Compared with normal group, the cardiac myocyte morphologe were not clearly, cytoplasmic swelling, inflammatory cells with in the visible. After the treatment with PSP which the situation gradually improved in some extent;Compared with normal group, the myocardial tissue of TLR4, NF-?B and MIF the protein expressions increased significantly (P<0.01); contrast with diabetes group, the myocardial tissue of TLR4, NF-?B and MIF in PSP 250mg/kg,500mg/kg and 1000mg/kg dose groups the protein expressions were reduced (P<0.05 or P<0.01);ConclusionsPSP can depress blood glucose, blood lipids and improve heart function, reduce myocardial inflammation injury, inhibit the expression of inflammatory cytokines; PSP has a protective effect on myocardial inflammation in diabetic rats, the possible mechanism is inhibition the expression of TLR4, NF-?B; MIF.
