The effect of Xiaoyao Powder on follicular maldevelopment with liver depression and spleen deficiency syndrome induced by chronic stress andthe research on Ovarian local regulation factor BDNF.Objective Firstly, observing the effect of Xiaoyao Powder on follicular maldevelopment with liver depression and spleen deficiency syndrome induced and clarifying the effect of Xiaoyao Powder on expression of Ovarian local regulation factor BDNF by chronic stress.Then, intensively studying how Xiaoyao Powder improving follicular maldevelopment by chronic stress and providing the basis for mechanism of Ovarian local regulation factor BDNF.Methods 60 healthy SPF grade ICR female mice are randomly divided into 3 group(20 mice per group): blank control group, chronic stress group and Xiaoyao powder group. Liver depression and spleen deficiency syndrome was induced by chronic unpredictable mild stress. The weight of mice was measured to analysis the changes of body weight under chronic stress state. Ovulation was judged by the total number of oocytes ovulated into ampulla of fallopian tube after treatment with gonadotropin. The different developmental stages of follicle were observed by HE staining. The number of ovulation and the row of the blastocyst formation rate was analyzed to understand the developmental potential of follicular and the development potential of oocyte. Ovarian endocrine was judged by plasma E2 and progesterone level using ELISA method. The effect of Xiaoyao Powder on expression of Ovarian local regulation factor BDNF by chronic stress was tested by Western blot method.Results1. The influence of body weight on mice: In the seventh day of the stress, when compared with blank control group, there was no significant difference(P > 0.05) for both chronic stress group and xiaoyao powder group in terms of mice body weight. In the fourteenth days of the stress, the difference was statistically significant(P < 0.01) between chronic stress group and blank control group in terms of mice body weight. In the twenty-first days of the stress, when Chronic stress group was compared with blank control group, the difference of weight of mice was statistically significant(P < 0.01); when xiaoyao powder group was compared with blank control group, the difference of mice body weight was statistically significant(P < 0.01); when Chronic stress group was compared with xiaoyao powder group, the difference of mice body weight was statistically significant(P < 0.01).2. The influence of xiaoyao powder on follicles of the chronic stress mice: The total number of ovulated oocytes in mice reduced after chronic stress(P<0.05); The total number of ovulated oocytes was markedly increased in xiaoyao powder compared with Chronic stress group(P<0.05).The number and the form of antral follicle in blank control group were normal. The number of normal secondary and antral follicle significantly decreased(P<0.05)and the number of atretic secondary and antral follicle significantly increased(P<0.05) of Chronic stress group. The number of normal secondary and antral follicle was markedly increased in xiaoyao powder compared with Chronic stress group(P<0.05), and the number of atretic secondary and antral follicle significantly increased in xiaoyao powder compared with Chronic stress group(P<0.05).3.Compared with blank control group, the formation rate of blastocyst significantly decreased(P<0.001)in chronic stress group, Compared with chronic stress group, the formation rate of blastocyst significantly increased(P<0.001)in xiaoyao powder.4. Effects of estradiol and progesterone levels: Compared with blank control group, the plasma level of E2 and progesterone significantly decreased(P<0.05)in chronic stress group, compared with chronic stress group, the plasma level of E2 and progesterone significantly increased(P<0.05)in xiaoyao powder.5. Immunoblotting assays the effect of Xiaoyao Powder on expression of Ovarian local regulation factor BDNF by chronic stress: Compared with blank control group, chronic stress group decreased the expression of Western blotting(P<0.05). Compared with chronic stress group, Xiaoyao Powder increased expression of Western blotting(P<0.05).Conclusion1. Xiaoyao powder could improve ovulation, follicular development and reproductive endocrine in mice with liver depression and spleen deficiency syndrome induced by chronic stress.2. Xiaoyao powder could regulate the expression of BDNF, promote the developmental potential of follicular and the development potential of oocyte.
