Objective:Through replicating animal model of vascular dementia,for moxibustion treat model rats”Da zhui”?”Bai hui”?”Shen ting”point,after treatment for 3 courses,diving platform experiment test results of learning and memory, combined with the neurobehavioral scores, before and after treatment comparison and contrast between groups, verify the effect of moxibustion treatment of vascular dementia, obtain evidence of moxibustion therapeutic effect. Through the moxibustion treatment of vascular dementia rats and carries on the contrast between groups,after treatment for 3courses,Intraperitoneal injection of Brd U fluid,Brd U immune double standard combined with confocal laser technology to track of NSCs differentiation, migration and other specific protein expression of the process,and mature endothelial cells, homing,secretionprocess,to obtain evidence of the moxibustion promote "neurogenesis";Brd U/CD34 expression of double testing new neurons peripheral vascular density,obtaining moxibustion promote "neurogenesis" also promotes "angiogenesis".Methods:Experiment 1:Selects the weight of 250 ~ 300 g clean level 80 adult male Wistar rats,The diving platform experiments for screening, random separated only 8 animals in front of the building as a false model group(control group), the rest by screening qualified rats four vessels used to block(4- vessle occlusion, 4 vo) method system of city animal model of vascular dementia and the behavioral memory performance testing,combined with neurobehavioral scores, validation building success. Randomly selected from building a successful animal only makes moxibustion research on the effects of cognitive function of VD model rats, then 24 model rats were randomly divided into 8western medicine group, moxibustion group, model group, 8, and compared with model control group. moxibustion group in the animal building began to four days after treatment, Moxibustion treatment of VD rats”Da zhui”?”Bai hui”?”Shen ting”point, 1times a day, 14 times for a period of treatment, a course of 7 d after the rest, treatment 3procedures; the western medicine group was given nootropic piracetam solution to fill the stomach, 2 times a day, course same as moxibustion group; model group with moxibustion group on time is the same,only do rat board fixed and do not make any other treatment;false model control group do not make any related treatment. each group of rats after treatment for 3 courses,the diving platform test results of learning and memory, combined with the neurobehavioral scores, contrast between groups, before and after treatment comparison to verify the curative effect of moxibustion.Experiment 2: Made of VD model method with experiment 1, From building a successful animals were used in the moxibustion randomly selected 18 rats nerve of VD model rats- vascular research on the effects of symbiosis, and then rats were divided into 3 groups according to random number method, moxibustion group, western medicine group, the VD model group, each group of six. Treatment with experiment 1,each group of rats in 24 h after completion of three course to be put to death, within 12 h before death, intraperitoneal injection of every 4 h Brd U 50 mg/kg, a total of three times,to Brd U tag,animals in the last 4h after injection of Brd U, with 6% chloral hydrate(350mg/kg) for abdominal cavity depth of anesthesia, reoccupy heparinization saline solution and paraformaldehyde solution for materials of cerebral perfusion(brain), after production, dyeing using laser confocal microscope combined with Brd U expression detection NSCs proliferation, differentiation and mature neurons, Brd U double express detect peripheral vascular density of new neurons, the moxibustion promote neurogenesis in VD rats, and neurogenesis, "angiogenesis" compliance.All the data used in experiment 1 and experiment 2 SPSS12.0 statistics software for processing, the standard deviation of measurement data are used to mean ±said( sx ±), take P<0.05 as was statistically significant, P<0.01 for significant statistical significance.Results:(1)The grades of learning and memory and neurobehavioral scores:Model group compared with false model group, rats learning and memory performance of error number increases, the incubation period shorten, neurobehavioral scores improved(P<0.01);That building success; Groups of rats after treatment for 1 ~ 2 period of treatment, western medicine group, the moxibustion group compared with model group,the rats learning and memory score of the wrong number, the incubation period and neurobehavioral scores were significantly different(P<0.01).The index difference compared with the western medicine group moxibustion group had no statistical significance(P>0.05);Groups of rats after the third treatment after the treatment,western medicine group, the moxibustion group compared with model group, the rats learning and memory score of the wrong number, the incubation period and neurobehavioral scores were significantly different(P<0.01).Compared with the western medicine group, moxibustion group of each index value was statistically significant(P<0.05). It shows that: moxibustion, western medicine in the VD model rats have a significant effect on improve cognitive function, and the curative effect of moxibustion is superior to western medicine treatment.(3)VD rats ischemic hippocampus Brd U/Nestin, Brd U/Neu N double dye cell count:After 3 courses of treatment, western medicine, moxibustion group compared with model group, Brd U/Nestin and Brd U/Neu N double dye the expression of positive cells increased obviously(P<0.01),In the treatment of the same period of moxibustion group Brd U/Nestin and Brd U/Neu N double positive cells expression is superior to western medicine group(P<0.05).This shows that: after three treatments after the treatment, moxibustion, western medicine in the treatment of VD model rats on the ischemic hippocampus the proliferation and differentiation of neural stem cells have a very good promoting effect, and the curative effect of moxibustion than western medicine curative effect is prominent.(4)VD rats ischemic hippocampus Brd U/CD34 and microvascular density: After 3courses of treatment, compared with VD model group,western medicine, moxibustion group of double standard positive Brd U/CD34 expression and microvascular density MVD are increasing(P<0.05);Compared with the western medicine group, moxibustion group of double standard positive Brd U/CD34 expression and microvascular density MVD are increased(P<0.05). It shows that: after 3 courses of treatment, moxibustion and the method of western medicine can increase the VD model rats ischemic hippocampus microvascular density, thereby promoting angiogenesis, and moxibustion curative effect is superior to western medicine curative effect.Conclusion:(1)Moxibustion can improve cognitive function in VD rats, reduce the neurobehavioral score, is a kind of effective means for the treatment of VD;(2)Moxibustion can improve the VD rats ischemic hippocampus Brd U/Nestin,Brd U/Neu N double standard the expression of positive cells, promote the proliferation and differentiation of neural stem cells after cerebral ischemia, contribute to restoration of nerve function.(3)Moxibustion can improve the VD rats ischemic hippocampus Brd U/CD34 positive double standard cells, increase the microvascular density, promote angiogenesis.
